Endothelial Differentiation of Mouse Mesenchymal Stem Cells

The purpose of this study focused on the isolation ofmesenchymal stem cells from bone marrow, in vitro cultivation and morphology assessment of isolated cells, morphological and phenotypic characterization and evaluation of multilinearity capability through differentiation of primaryculture towards endothelial line. The biological material was represented byCD1 mice that were killed and the femur bone was collected under maximumsterility conditions. The cultivation of mesenchymal stem cells isolated frombone marrow was performed using the IMDM medium, supplemented with 10% FCS, 1% Antibiotic-Antimycotic, 1%glutamine, 1% NEA and 5% horse serum. Mesenchymal stem cell differentiation towards the endothelial line was induced using the EBM2 medium supplementedwith EGM2 SingleQuots. The multilinearity of murine mesenchymal stem cells was demonstrated by flow cytometry, after differentiation, using anti CD34 andCD105 (Endoglin) antibodies. The results proved that the EBM2 treated cultures supplemented with SingleQuots showed specific endothelial morphologic characteristics, 4% of them being positive for CD34, while 67.2 % were positive for CD105.
