    摘要:Lyme disease has increased both in incidence and geographic extent in the United States and Canada over the past two decades. One of the underlying causes is changes during the same time period in the distribution and abundance of the primary vectors: Ixodes scapularis Say and Ixodes pacificus Cooley and Kohls in eastern and western North America, respectively. Aside from short periods of time when they are feeding on hosts, these ticks exist in the environment where temperature and relative humidity directly affect their development, survival, and host-seeking behavior. Other important factors that strongly influence tick abundance as well as the proportion of ticks infected with the Lyme disease spirochete, Borrelia burgdorferi, include the abundance of hosts for the ticks and the capacity of tick hosts to serve as B. burgdorferi reservoirs. Here, we explore the linkages between climate variation and: 1) duration of the seasonal period and the timing of peak activity; 2) geographic tick distributions and local abundance; 3) enzootic B. burgdorferi transmission cycles; and 4) Lyme disease cases. We conclude that meteorological variables are most influential in determining host-seeking phenology and development, but, while remaining important cofactors, additional variables become critical when exploring geographic distribution and local abundance of ticks, enzootic transmission of B. burgdorferi, and Lyme disease case occurrence. Finally, we review climate change-driven projections for future impact on vector ticks and Lyme disease and discuss knowledge gaps and research needs.

    摘要:Unblocked fleas can transmit Yersinia pestis, the bacterium that causes plague, shortly (≤4d) after taking an infectious bloodmeal. Investigators have measured so-called early-phase transmission (EPT) efficiency in various fleas following infection with highly bacteremic blood (≥108cfu/ml). To date, no one has determined the lower limit of bacteremia required for fleas to acquire and transmit infection by EPT, though knowing this threshold is central to determining the length of time a host may be infectious to feeding fleas. Here, we evaluate the ability of Oropsylla montana (Baker) to acquire and transmit Y. pestis after feeding on blood containing 103 to 109cfu/ ml. We evaluated the resulting infection prevalence, bacterial loads, and transmission efficiency within the early-phase time period at 1d postinfection. Fleas acquired infection from bacteremic blood across a wide range of concentrations, but transmission was observed only when fleas ingested highly bacteremic blood.

    摘要:The pruritic skin disease scabies is caused by the burrowing of the itch mite Sarcoptes scabiei (De Geer). It is difficult to diagnose this disease because its symptoms often resemble those of other skin diseases. No reliable blood or molecular diagnostic test is available. The aim of this project was to begin to characterize the scabies proteome to identify scabies mite proteins, including those that may be useful in the development of a diagnostic test or vaccine. Various scabies mite extracts were separated by two-dimensional electrophoresis, and 844 Coomassie Blue-stained protein spots were excised, subjected to trypsin digestion, and analyzed by Matrix Assisted Laser Desorption/Ionization Time-Of-Flight/Time-Of-Flight (MALDI-TOF/TOF) mass spectrometry (MS). Tryptic fragment sequences determined by MS were searched against the recently completed S. scabiei annotated genome, leading to the identification of >150 proteins. Only 10 proteins hit to previously identified scabies proteins including actin, tropomyosin, and several ABC transporters. Thirteen proteins had homology to dust mite allergens (members of groups 8, 10, 13, 17, 20, 25, and 28). Most other sequences showed some homology to proteins in other mites and ticks including homologs of calmodulin, calreticulin, lipocalin, and glutathione-S-transferase. These data will now allow the identification of the proteins to which scabies patients produce antibodies, including those that may be good candidates for inclusion in a diagnostic test and vaccine.

    摘要:Knockdown resistance ( kdr ) in insects resulting from mutation(s) in the voltage-gated sodium channel (VGSC) gene is one of the mechanisms of resistance against DDT and the pyrethroid group of insecticides. Earlier, we reported the presence of two classic kdr mutations, i.e., L1014F and L1014S in Anopheles stephensi Liston, a major Indian malaria vector affecting mainly urban areas. This report presents the distribution of these alleles in different An. stephensi populations. Seven populations of An. stephensi from six states of India were screened for the presence of two alternative kdr mutations L1014F and L1014S using allele-specific polymerase chain reaction assays. We recorded the presence of both kdr mutations in northern Indian populations (Alwar and Gurgaon), with the preponderance of L1014S, whereas only L1014F was present in Raipur (central India) and Chennai (southern India). None of the kdrmutations were found in Ranchi in eastern India and in Mangaluru and Mysuru in southern India. This study provides evidence for a focal pattern of distribution ofkdralleles in India.

    摘要:The endosymbiotic bacterium Wolbachia pipientis infects many species of insects and has been transinfected into the mosquito Aedes aegypti (L.), the primary vector of dengue virus (DENV). Recently, it has been shown that Wolbachia blocks the replication and transmission of RNA viruses, such as DENV, in a number of mosquito species including Ae. aegypti and Aedes albopictus (Skuse), which is naturally infected with Wolbachia and considered a secondary vector for DENV. The mosquito species Aedes notoscriptus (Skuse) is highly prevalent in Australia, including in areas where DENV outbreaks have been recorded. The mosquito has been implicated in the transmission of Ross River and Barmah Forest viruses, but not DENV. We investigated whether Wolbachianaturally infects this mosquito species and whether it has an impact on the ability ofAe. notoscriptusto transmit DENV. We show, for the first time, thatAe. notoscriptusis naturally infected with a strain ofWolbachiathat belongs to supergroup B and is localized only in the ovaries. However,Wolbachiainfection inAe. notoscriptusdid not induce resistance to DENV and had no effect on overall DENV infection rate or titer. The presence of a nativeWolbachiainAe. notoscriptuscannot explain why this mosquito is an ineffective vector of DENV.

    摘要:Field studies of the ecology of eastern equine encephalitis virus (EEEV; family Togaviridae , genus Alphavirus ) in the southeastern United States have demonstrated that Culex erraticus (Dyar and Knab) is the most common mosquito at many enzootic sites and is often infected with the virus. However, the competence of Cx. erraticus for EEEV has not been explored in detail. Culex erraticus females were collected from the field and fed upon EEEV-infected chicks. The infected mosquitoes were provided honey for nutrition and to monitor for time to infectiveness. Of the mosquitoes that survived the 14-d postfeeding period, 89% were infected and 84% had evidence of a disseminated infection, though titers were generally low. EEEV was first detected in honey 6 d postinfection and was detected in samples collected from 94% of the mosquitoes with a disseminated infection overall. These data and others were then employed to estimate the relative vectorial capacity of Cx. erraticus at an EEEV enzootic site in Alabama. The vectorial capacity of Cx. erraticus at this site was 44% of Culiseta melanura(Coquillett), the accepted enzootic vector, suggestingCx. erraticusmay play a role in transmitting EEEV in areas where it is abundant andCs. melanurarare.

    摘要:Single and mixed species densities of adult Aedes albopictus and Culex quinquefasciatus were manipulated to determine if different combinations affected their egg laying preference or mortality rates. Oviposition was measured in environments that contained containers of different surface areas (small cups vs. larger bowls), and the number of eggs ( Aedes ) and egg rafts or larvae ( Culex ) deposited by each species was examined with respect to intra- and interspecific density treatment levels. Mixed species densities did not have an effect on survivorship, but single species densities did affect longevity, with higher densities leading to shorter life spans in Cx. quinquefasciatus : Cx. quinquefasciatus lived longer than Ae. albopictus overall. There was no significant effect of density combinations on oviposition patterns in either species, but Cx. quinquefasciatuslaid more rafts in bowls compared with cups. There is little evidence of adult interactions between these species; however, future experimental work is necessary to more fully characterize the possible effects of adult interactions on these species.

    摘要:Aedes albopictus (Skuse) is a container-breeding species with considerable public health importance. To date, Ae. albopictus oviposition behavior has been assessed in outdoor conditions, but only with laboratory-reared specimens. In outdoor large-cage and field studies, we used an attractive self-marking ovipositional device to assess Ae. albopictus skip oviposition behavior. In field studies, 37 wild Ae. albopictus that visited an attractive self-marking ovisite were subsequently captured at a sticky ovitrap within a 4-d period. Because the average Ae. albopictus gonotrophic period is 4.5–6 d, the wild-caught Ae. albopictus visited at least two oviposition sites within a single gonotrophic period. This provided field-based indirect evidence of skip oviposition. The mean distance traveled (MDT) during the 20-d evaluations ranged from 58 to 78 m. The maximum observed distance traveled was 149 m, which was the outer edge of our trapping ability. As populations of Ae. albopictus increased, the MDT during the 4- and 20-d post-marking period increased significantly. Additional observations of wild-marked and captured Aedes triseriatus (Say) are discussed.

    摘要:Insect-specific flaviviruses (ISFVs) commonly infect vectors of mosquito-borne arboviruses. To investigate whether infection with an ISFV might affect mosquito flight behavior, we quantified flight behavior in Culex pipiens L. naturally infected with Culex flavivirus (CxFV). We observed a significant reduction in the scotophase (dark hours) flight activity of CxFV-positive mosquitoes relative to CxFV-negative mosquitoes, but only a marginal reduction in photophase (light hours) flight activity, and no change in the circadian pattern of flight activity. These results suggest that CxFV infection alters the flight activity of naturally infected Cx. pipiens most dramatically when these vectors are likely to be host seeking and may therefore affect the transmission of medically important arboviruses.

    摘要:Temperature–food interactions in the larval environment can affect life history and population growth of container mosquitoes Aedes aegypti (L.) and Aedes albopictus Skuse, the primary vectors of chikungunya and dengue viruses. We used Ae. aegypti, Ae. albopictus, and dengue-1 virus (DENV-1) from Florida to investigate whether larval rearing temperature can alter the effects of larval food levels on Ae. aegypti and Ae. albopictus life history and DENV-1 infection and vertical transmission. Although we found no effect of larval treatments on survivorship to adulthood, DENV-1 titer, or DENV-1 vertical transmission, rates of vertical transmission up to 16–24% were observed in Ae. albopictus and Ae. aegypti, which may contribute to maintenance of this virus in nature. Larval treatments had no effect on number of progeny and DENV-1 infection in Ae. aegypti, but the interaction between temperature and food affected number of progeny and DENV-1 infection of the female Ae. albopictus parent. The cooler temperature (24°C) yielded the most progeny and this effect was accentuated by high food relative to the other conditions. Low and high food led to the highest (∼90%) and lowest (∼65%) parental infection at the cooler temperature, respectively, whereas intermediate infection rates (∼75–80%) were observed for all food conditions at the elevated temperature. These results suggest that temperature and food availability have minimal influence on rate of vertical transmission and a stronger influence on adults of Ae. albopictus than of Ae. aegypti, which could have consequences for dengue virus epidemiology.

    摘要:Evolutionary theory predicts that vector-borne pathogens should have low virulence for their vector because of selection against pathogens that harm the vector sufficiently to reduce transmission. Environmental factors such as nutritional stress can alter vector–pathogen associations by making the vectors more susceptible to pathogens (condition-dependent competence) and vulnerable to the harm caused by pathogen replication (condition-dependent virulence). We tested the hypotheses of condition-dependent competence and condition-dependent virulence by examining the interactive effects of short-term sugar deprivation and exposure to La Crosse virus (LACV) in female Aedes albopictus (Skuse). We predicted that infection status interacts with sugar deprivation to alter willingness to blood feed and fecundity in the second gonotrophic cycle (condition-dependent virulence). Sugar deprivation had no effect on body infection or disseminated infection rates. Infection status, sugar treatment, and their interaction had no effect on fecundity. Mosquitoes that had intermittent access to sugar were significantly more willing to take a second bloodmeal compared with those that had continuous access to sugar. Infection status and the interaction with sugar treatment had no effect on blood-feeding behavior. Thus, we found no evidence of short-term sugar deprivation leading to condition-dependent competence for, or condition-dependent virulence of, LACV in Ae. albopictus.

We infected laboratory-reared P. sergenti originating from Turkey and Israel to compare their susceptibility to L. tropica. In both tested groups, heavy late-stage infections with the presence of metacyclic forms and colonization of the stomodeal valve were observed. The similar development of Leishmania in both sand fly colonies indicates that the different geographical origin of P. sergenti is not reflected by a different vector competence to L. tropica. Additionally, we tested the effect of the gregarine Psychodiella sergenti on L. tropica coinfections; no apparent differences were found between P. sergenti infected or not infected by gregarines.

    摘要:Fourth instars of Culex quinquefasciatus (Say) (Diptera: Culicidae) were selected with a recombinant bacterial strain synthesizing the mosquitocidal proteins from Lysinibacillus sphaericus (Bin) and Cry11Ba and Cyt1Aa from Bacillus thuringiensis. Selection was initiated in Generation 1 with a concentration of 0.04 μg/ml, which rose to a maximum selection concentration of 8.0 μg/ml in Generation 14, followed by an unexpected, rapid increase in mortality in Generation 15. Subsequently, a selection concentration of 0.8 μg/ml was determined to be survivable. During this same period, resistance rose to nearly 1,000-fold (by Generation 12) and declined to 18.8-fold in Generation 19. Resistance remained low and fluctuated between 5.3 and 7.3 up to Generation 66. The cross-resistance patterns and interactions among the component proteins were analyzed to identify possible causes of this unusual pattern of evolution. Poor activity in the mid-range concentrations and lower-than-expected synergistic interactions were identified as potential sources of the early resistance. These findings should be considered in the development of genetically engineered strains intended to control nuisance and vector mosquitoes.

    摘要:The human head louse is a cosmopolitan ectoparasite and frequently infests many people, particularly school-age children. Due to widespread pyrethroid resistance and the lack of efficient resistance management, there has been a considerable interest in the protection of uninfested people and prevention of reinfestation by disrupting lice transfer. In this study, two nonclinical model systems (in vitro and in vivo) were used to determine the efficacy of the infestation deterrents, Elimax lotion and Elimax shampoo, against human head lice or poultry chewing lice, respectively. With in vitro assessments, female head lice exhibited significantly higher avoidance responses to hair tufts treated with either of the test formulations, which led to significantly higher ovipositional avoidance when compared with female lice on control hair tufts. Additionally, both formulations were determined to be competent infestation deterrents in a competitive avoidance test in the presence of a known attractant (head louse feces extract). In in vivo assessments using a previously validated poultry model, Elimax shampoo was determined to be an efficacious deterrent against poultry chewing lice within Menopon spp. and Menacanthus spp.

Males of D. filippovae may be distinguished from those of D. auratus, D. compactus, and D. limbooliati by the following suite of characters: conscutum broadly oval with convex lateral sides, widest posteriorly; trapezium-like shape of pseudoscutum; central brown patch on conscutum indistinct; coxa I with internal spur narrowly triangular with tapering apex and external spur fairly long, narrowly triangular with tapering apex; numerous internal spurs on coxa IV; and trochanter I with moderate, broadly triangular spur with tapering apex. Females of D. filippovae may be distinguished from those of D. auratus, D. compactus, and D. limbooliati by the following suite of characters: central patch of scutum narrow, distinct line interrupted around midlength; more rounded shape of scutum; long and distinct alloscutal setae; moderately broad U-shaped genital aperture with preatrial fold bulging anteriorly and thereafter sharply sloping to flat surface posteriorly; coxa I with internal spur narrowly triangular with tapering apex and external spur fairly long, narrowly triangular with tapering apex; and trochanter I with moderate, broadly triangular spur with tapering apex. Dermacentor filippovae is known from Thailand and Vietnam where the adults were collected from wild boar, Sus scrofa L., and vegetation.

    摘要:Homoplasmy, the occurrence of a single mitochondrial DNA haplotype within an individual, has been the accepted condition across most organisms in the animal kingdom. In recent years, a number of exceptions to this rule have been reported, largely due to the ease with which single nucleotide polymorphisms can be detected. Evidence of heteroplasmy—two or more mitochondrial variants within a single individual—has now been documented in a number of invertebrates; however, when present, heteroplasmy usually occurs at low frequencies both within individuals and within populations. The implications of heteroplasmy may be far reaching, both to the individual in relation to its health and fitness, and when considering the evolutionary dynamics of populations. We present novel evidence for frequent mtDNA heteroplasmy in the bed bug, Cimex lectularius L. (Hemiptera: Cimicidae). Our findings show that heteroplasmy is common, with 5 of 29 (17%) populations screened exhibiting two mitochondrial variants in a ∼1:2 ratio within each individual. We hypothesize that the mechanism underlying heteroplasmy in bed bugs is paternal leakage because some haplotypes were shared among unrelated populations and no evidence for nuclear mitochondrial DNA sequences was detected.

    摘要:Dengue vector Aedes aegypti L. is invading peri-urban and rural areas throughout Latin America. Our previous research in the Peruvian Amazon has shown that river boats are heavily infested with immature and adult Ae. aegypti mosquitoes, likely playing a major role in their long-distance dispersal and successful invasion. However, the presence of immature mosquitoes provides no information about the timing of oviposition, and whether it took place in the boats. Here, we used baited ovitraps deployed on river boats to test the hypothesis that Ae. aegypti oviposition occurs during boat travel. We deployed 360 ovitraps on 60 different barges during August and October of 2013, and February 2014 (with 20 barges sampled during each month). We found that Ae. aegypti mosquitoes in 22 individual ovitraps from 15 of the 60 barges (premise index 25%) across all sampling dates. Further, the distribution of Ae. aegypti egg abundance was highly aggregated: 2.6% of traps (N = 7) were responsible for 71.8% of eggs found, and 1.5% of traps (N = 4) were responsible for all (100%) of the larvae found. Similarly, 5% of boats were responsible for the 71.47% of eggs. Our results provide strong evidence that Ae. aegypti oviposition commonly occurs during boat travel. Baited ovitraps could represent a cost-effective means of monitoring and controlling mosquito populations on boats.

    摘要:Re-examination of Dermacentor taiwanensis Sugimoto, 1935 specimens in the United States National Tick Collection revealed that two morphologically distinct Dermacentor species were identified under this name. One of them corresponds to Sugimoto’s description of D. taiwanensis, while another species is identical to Schulze’s Dermacentor bellulus (Schulze, 1935). The latter species has not been considered valid by recent workers. D. bellulus is reinstated here as a valid species and all its stages are redescribed. The adults of D. taiwanensis are also redescribed, and its immature stages are described for the first time. Males and females of D. bellulus can be distinguished from those of D. taiwanensis by the shape of the conscutum and scutum, color pattern, genital structures, size of the palpi and cornua, and the spurs of coxa I. Nymphs of D. bellulus can be distinguished from those of D. taiwanensis by the shape of the scutum, basis capituli, and the hypostomal dentition. Larvae of D. bellulus can be differentiated from those of D. taiwanensis by the shape of the basis capituli, and the degree of development of the auriculae and spur on palpal segment III ventrally. D. bellulus has been recorded from China, Japan, Nepal, Taiwan, and Vietnam; adults have been collected from wild boars, bears, panda, dog, and human; the immature stages are known from rodents, hares, ferret-badger, and bamboo-partridge. D. taiwanensis is found in China, Taiwan, and Vietnam; adults have been collected from wild boars; the immature stages are known from rodents, hares, mustelids, and domestic dog.
