    摘要:Bovine respiratory disease (BRD) is the most common cause of morbidity and mortality in North American beef cattle. Mannheimia haemolytica is the bacterial pathogen most often isolated from cattle with BRD, and the prevalence of antimicrobial resistance (AMR) in this organism has increased in recent years. Antimicrobials are commonly used to prevent BRD in cattle at high risk of developing BRD; however, recent work would suggest that this practice might be one factor contributing to the increased prevalence of AMR in M. haemolytica. We hypothesized that the administration of the short-acting fluoroquinolone, enrofloxacin, would be just as effective as the long-acting triamilide, tulathromycin, in preventing BRD but would be less likely to select for AMR M. haemolytica in stocker calves at high risk of developing BRD. Three hundred forty-one stocker calves were enrolled in the study with 172 calves in 4 pens being randomly assigned to treatment with enrofloxacin and 169 calves in 4 pens randomly assigned to treatment with tulathromycin. Calves within each treatment group were allocated to one of 4 replicate pens based on the week of enrollment. Of calves receiving enrofloxacin, 33.7% required treatment for BRD at least once within 45 d after arrival, compared with 18.3% of calves receiving tulathromycin (P = 0.040). The percentages of calves that required more than one treatment for BRD within 45 d after arrival did not differ statistically for those receiving enrofloxacin compared with those receiving tulathromycin (10.5% and 4.7%, respectively; P = 0.107) Likewise, the percentages of calves that died during the 45-d follow-up period did not differ for those receiving enrofloxacin compared with those receiving tulathromycin (12.2% and 10.1%, respectively; P = 0.592). Mannheimia haemolytica was cultured from 11% of calves sampled at arrival and from 50% of calves sampled at revaccination 14 to 17 d later. There was a significanst effect of sampling time on the proportion of calves carrying multidrug-resistant (MDR) isolates, with calves having a higher prevalence of MDR isolates at revaccination than arrival (100% vs. 13%; P < 0.001). Future research evaluating the impact of MDR on response to antimicrobial therapy is necessary.

    摘要:In Western Canada, approximately half of the calves produced are castrated before 1 wk of age. Therefore, it is important to identify effective analgesic drugs to mitigate pain associated with castration and consequently improve animal welfare. The aim of this study was to assess the efficacy of a single s.c. dose of meloxicam at mitigating pain associated with knife and band castration in 1-wk-old calves. Seventy-two Angus crossbred bull calves (47.3 ± 6.70 kg of body weight (BW), 1 wk old) were used in a 3 × 2 factorial design where main factors included castration method–sham (CT), band (BA) or knife (KN) castration– and medication–lactate ringer (NM) or 0.5 mg/kg BW of meloxicam (M). Measurements included different physiological and behavioral parameters. Samples were collected on day −1, immediately before castration (T0); and 60, 90, and 120 min and 1, 2, 3, and 7 d after castration except for visual analog scale (VAS) which was collected at the time of castration. The salivary cortisol concentrations were greater (P = 0.04) in KN and BA calves than CT calves 60 min after castration, while 90 min after castration BA had greater concentrations than CT calves. Substance P concentrations were greater (P = 0.04) in NM calves than M calves on d 3 and 7 after castration. The serum amyloid-A (SAA) concentrations were greater (P = 0.05) in KN calves than BA and CT calves on days 0, 2, and 3, while BA calves had greater SAA concentrations on day 7 than KN and CT calves. The visual analog scores were greater (P < 0.01) in KN calves than BA, and in BA compared to CT calves. The KN calves tail flicked more (P < 0.01) than BA and CT calves, and NM calves tail flicked more (P = 0.03) than M calves. No castration or medication effect (P > 0.10) was observed for stride length, walking, standing, lying ventral, eating, foot stamping, head turning, lying and standing percentage, performance, platelets, or body temperature. Overall, knife castrated calves exhibited a greater acute pain response than band castrated calves. Meloxicam was able to reduce substance P concentrations, white blood cell counts and number of tail flicks after castration, suggesting that the drug was able to mitigate acute pain to some extent. However, meloxicam did not have an effect on the other physiological and behavioral parameters assessed.

    摘要:Bovine respiratory disease complex (BRDC) is a multifactor disease, and disease incidence may be associated with an animal’s commensal bacterial populations (microbiome) in the upper nasal cavity. Identifying these commensal bacterial populations in the upper nasal cavity may help us to understand the impact of the microbiome on incidence of BRDC in cattle. Various sampling techniques have previously been utilized to evaluate the microbiome of different locations of the upper nasal cavity in cattle. Therefore, our objective was to determine whether bacterial populations of the nasal cavity vary based on these sampling locations. Two common sampling techniques were evaluated, including 6-inch nasal swabs and deep nasopharyngeal swabs. Nasal swabs from calves were collected when the animal was diagnosed with BRDC after weaning in the feedlot in addition to collection of samples from asymptomatic cohorts. Samples were pooled in groups based on year the animal was in the feedlot (2015 or 2016), when the animal was diagnosed with BRDC (1 to 5 weeks after weaning), type of sample (6-inch nasal swab or deep nasopharyngeal swab), and health status (diagnosis with BRDC or control). Variable regions 1 through 3 along the 16S rRNA gene were amplified by PCR and sequenced using next-generation sequencing (Illumina MiSeq) for identification of the bacterial taxa present. Overall, sampling site did not consistently influence diversity of the bacterial populations of the upper nasal cavity. However, the effect of disease incidence on the microbiome was depended on sampling time after weaning (P = 0.0462) for 2015, while the main effects of sampling time after weaning (P = 0.00992) and disease phenotype (P = 0.012) were significant for 2016. These data for 2016 demonstrate that in addition to bacterial profiles changing throughout weaning, calves diagnosed with BRDC have different bacterial profiles compared to their control cohorts. In addition, evaluation of the microbiome identified predominant bacteria genera in the upper nasal cavity included those previously reported to be associated with cattle diagnosed with BRDC including Mycoplasma sp., Psychrobacter sp., and Mannheimia sp. In summary, these results demonstrate that shorter, less invasive 6-inch nasal swabs produce similar results to deep nasopharyngeal swabs.

    摘要:In this review, the potential for use of soy-derived bioactive compounds as immunomodulatory feed additives in pigs is discussed. Soy is a major component of the modern U.S. swine diet in today’s commercial industry, providing the bulk of dietary AA necessary for growth and production. However, soy use has generally been limited in early growth phases, during which the risks of immunological insult and disease are among the highest. Improvements of soybean processing and development of soy protein products with little to no antinutritional factors have made soy more appropriate for use in young pigs but additional processing may affect bioactive compound levels in the feed. The bioactive compounds of interest for this review are soy isoflavones and soy saponins. Soy isoflavones are flavonoid compounds with a range of biological activity including moderate estrogenic effects at low biological concentrations. Although estrogenic effects are of more interest in human medical research, isoflavones are also known for their anti-inflammatory, antioxidative properties at cellular levels, engaging several receptors and pathways including inhibition of NF-κB activation and inducible-nitric oxide synthase enzymes, thereby ascribing antiviral properties. Saponins, amphipathic glycoside compounds, also engage anti-inflammatory pathways, though their biological activity in pigs has not been well investigated and seem to mainly be observed on the mucous membrane in the gastrointestinal tract. Regarding use as an immunomodulatory feed additive, supplemental soy isoflavones have been shown to improve immunological status of pigs and produce mild improvements of growth performance under certain disease challenges including porcine reproductive and respiratory syndrome virus. Although more in vivo research in pigs is needed to fully understand biological activity of these compounds in the live animal, soy-derived bioactive compounds show great potential as a health promoting feed additive for the modern swine industry.

    摘要:It is known that physiological overproduction of nitric oxide (NO) contributes to oxidative stress and inflammation. Our published studies indicated that vitamin A (VA) reduces NO-induced oxidative stress in bovine mammary epithelial cells (BMECs) by increasing antioxidant enzyme activities. However, the precise mechanism is unclear. The present study was conducted to examine the protective effects of VA on NO-induced damage to BMECs in vitro using diethylenetriamine nitric oxide (DETA-NO) as the NO donor and to explore the intracellular signaling mechanisms of VA that involve nuclear factor erythroid 2-related factor (Nrf2) and nuclear factor kappa-B (NF-κB). Subconfluent BMECs were divided into 10 treatment groups with 6 replicates per treatment and were cultured with dimethyl sulfoxide (DMSO, vehicle negative control) or 0, 0.05, 0.1, 0.2, 0.5, 1, 2, 3, or 4 μg/mL of VA for 24 h and then incubated in the absence or presence of DETA-NO (1,000 μmol/liter) and VA for an additional 6 h. The results showed that exposure to DETA alone decreased cell proliferation compared with the negative control. Pretreatment with VA promoted the proliferation of BMECs, increased the activities of antioxidative enzymes including selenoprotein glutathione peroxidase (GPx) and thioredoxin reductase (TrxR) and their gene and protein expression but decreased NO and interleukin 1 (IL-1) contents in a quadratic manner (P < 0.05). In addition, the expression of mRNA and protein of factors that are related to NF-κB or Nrf2 signaling pathways in BMECs were regulated by VA in a quadratic dose-dependent manner; VA at a concentration of 1 μg/mL exhibited the strongest effect. Together, these results suggest that VA promotes antioxidant functions of BMECs by regulating the synthesis of selenoproteins including GPx and TrxR and by reducing concentrations of IL-1 and NO in vitro by modulating Nrf2 and NF-κB signaling pathways.

    摘要:An experiment was conducted to determine the effect of feeding finishing pigs a corn-soybean (CSBM) diet or a CSBM diet supplemented with 30% dried distillers grains with solubles (DDGS), in combination with or without a growth-promoting ionophore (0 or 30 mg narasin/kg of diet), has on manure composition, microbial ecology, and gas emissions. Two separate groups of 24 gilts (initial BW = 145.1 kg, SD = 7.8 kg) were allotted to individual metabolism crates that allowed for total but separate collection of feces and urine during the 48-d collection period. After each of the twice-daily feedings, feces and urine from each crate was collected and added to its assigned enclosed manure storage tank. Each tank contained an individual fan system that pulled a constant stream of air over the manure surface for 2 wk prior to air (day 52) and manure sampling (day 53). After manure sampling, the manure in the tanks was dumped and the tanks cleaned for the second group of pigs. Except for total manure Ca and P output as a percent of intake and for manure methane product rate and biochemical methane potential (P ≤ 0.08), there were no interactions between diet composition and narasin supplementation. Narasin supplementation resulted in increased manure C (P = 0.05), increased manure DM, C, S, Ca, and phosphorus as a percent of animal intake (P ≤ 0.07), and increased manure volatile solids and foaming capacity (P ≤ 0.09). No effect of narasin supplementation was noted on manure VFA concentrations or any of the gas emission parameters measured (P ≥ 0.29). In contrast, feeding finishing pigs a diet containing DDGS dramatically affected manure composition as indicated by increased concentration of DM, C, ammonia, N, and total and volatile solids (P = 0.01), increased manure DM, N, and C as a percent of animal intake (P = 0.01), increased manure total VFA and phenols (P ≤ 0.05), decreased gas emissions of ammonia and volatile sulfur compounds (VSC; P = 0.01), increased emissions of phenols and indoles (P ≤ 0.06), decreased methane production rate (P = 0.01), and slight differences in microbial ecology (R ≤ 0.47). In conclusion, feeding a diet which contains an elevated level of indigestible fiber (i.e., DDGS) resulted in more fiber in the manure which therefore dramatically affected manure composition, gas emissions, and microbial ecology, while narasin supplementation to the diet did not exhibit a significant effect on any of these parameters in the resultant swine manure.

    摘要:This study aimed to investigate the effect of supplemental yeast cell walls (YCW) on growth performance, gut mucosal glutathione pathway, proteolytic enzymes, and transporters in broiler chickens from 1 to 21 d of age. One-day-old broiler chickens (n = 480) were randomly allocated to 4 treatments with 6 replicates of 20 chicks each for diets containing YCW at 0, 0.5, 1.0, and 1.5 g/kg. The results showed that YCW supplementation increased (P < 0.05) ADFI and ADG by 15.3% and 16.0%, respectively, and the 2 higher doses of YCW had greater effects (P < 0.05) than the lower dose. For the glutathione pathway in the duodenal mucosa, the levels of reduced glutathione and glutathione reductase in the YCW treatments were increased (P < 0.05) by 15.6% and 17.4%, respectively, but glutathione S-transferases was not affected. Similarly, the YCW increased (P < 0.05) the mucosal activities of trypsin, dipeptidyl, and leucine aminopeptidase by 12.4%, 5.5%, and 17.3%, respectively, and the mRNA profiles of neutral, cationic, and oligopeptide transporters by 62.5%, 69.1%, and 11.5%, respectively. The YCW also increased (P < 0.05) the concentrations of Thr, Met, Ile, Leu, Lys, Arg, and Tyr in the blood by 8.8% to 39.2%. Additionally, the ADFI, ADG, reduced glutathione, trypsin, dipeptidyl aminopeptidase, leucine aminopeptidase, and cationic amino acid transporter increased linearly and quadratically (P < 0.05) with YCW supplementation. The results suggest that YCW supplementation can beneficially modulate intestinal glutathione pathway, proteolytic enzyme activity, and nutrient transport in growing animals.

    摘要:The objective of this study was to determine the chemical composition, DE and ME contents, and apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of AA in 7 cottonseed co-products fed to growing pigs. The 7 cottonseed co-products were: cottonseed meals (solvent extracted) with CP level of 46%, 50%, and 55% (46CSM, 50CSM, and 55CSM), cottonseed protein with CP level of 50% and 55% (50CSP and 55CSP), fermented cottonseed meal (CSMF), and expelled cottonseed meal (CSME). The DE and ME contents of the 7 test ingredients were measured using 48 crossbreed barrows (BW: 44.1 ± 4.2 kg) and 8 diets (including a corn-soybean meal basal diet) in a completely randomized design. The AID and SID of AA of the 7 test ingredients were determined using 7 crossbreed barrows (initial BW: 67.4 ± 8.5 kg) in a 6 × 8 Youden square design with 6 periods and 8 diets (including one N-free diet). The DE and ME values of the 7 cottonseed co-products varied from 12.72 to 15.63 MJ/kg and 12.24 to 14.83 MJ/kg, respectively. Among the 7 cottonseed co-products, 55CSP and CSME had the greatest (P < 0.05) ME and DE values compared to the other cottonseed co-products. In addition, 55CSP had the greatest (P < 0.05) AID and SID of all the AA tested except for Gly and Pro. In contrast, 46CSM or 55CSM had the lowest (P < 0.05) AID and SID of all the AA tested except for Gly and Pro. The 55CSP also had the greatest (P < 0.05) concentrations of standardized ileal digestible CP and all the AA tested except for Gly and Pro. In conclusion, the chemical composition, energy, and AA digestibility of cottonseed co-products with different processing techniques varied widely. Based on the energy and AA digestibility, cottonseed protein with CP level of 55% is a better dietary ingredient for growing pigs compared with the other cottonseed co-products.

    摘要:The objective of this study was to quantitatively summarize literature reporting endophyte-infected (Neotyphodium coenophialum) tall fescue (Festuca arundinacea) effects on cattle ADG. This meta-analysis evaluated endophyte infection level, climate, and forage yield using a literature dataset of 138 treatments from 20 articles. Three infection level measurements were tested: endophyte infection as a percentage of infected tillers (E%); ergovaline concentration in ppb ([E]); and total ergot alkaloid concentration ([TEA]). Three types of climate variables were used: base values (temperature, humidity, and relative humidity), climate indices (heat index and temperature-heat index [THI]), and novel climate variables accounting for duration of climate effects. Mixed effect models, weighted by 1/SEM, including a random effect of study were built for each factorial combination of measurement method and climate variable group. Because many studies were missing SEM, two datasets were used: one containing only data with SEM reported and one that also included missing-SEM data. For the complete-SEM dataset (CSD), models were weighted by 1/SEM. In the missing-SEM dataset (MSD) the mean reported 1/SEM was assigned as the weight for all missing SEM treatments. Although 18 initial models were created (2 × 3 × 3 factorial approach), the backward stepwise derivation resulted in models that included only endophyte infection level, suggesting a negative relationship between infection level and ADG. The CSD models predicted ADG to decrease 39 and 33 g/d with each increase of 100 ppb of [TEA] and [E], and by 39 g/d for each increase of 10% E%. In the MSD dataset, predicted ADG decreased by 39 and 33 g/d with each increase of 100 ppb of [TEA] and [E], and by 47 g/d for each increase of 10% E%. All relationships reported had P < 0.05. After visual inspection of the data, piecewise regression was used to identify an infection threshold (IT) of 60 ppb [E] and 11 E%, where the effect of infection level was constant on either side of the IT. The ADG was 40% and 49% greater for infection levels below the IT for [E] and E%, respectively. Across THI values in the analysis, ADG decreases ranged from 11.2% to 45.0% for cattle grazing endophyte-infected tall fescue compared to non-ergot alkaloid endophyte infected tall fescue. Pasture E%, [E], and [TEA] have a negative relationship with ADG in growing cattle, and increasing temperature decreases ADG when infection level is greater than the IT.

    摘要:The aim of the current study was to determine nutrient digestibility, VFA production, N metabolism, and CH4 production of canola (Brassica napus L.), rapeseed (B. napus L.), turnip (Brassica rapa L.), and annual ryegrass (Lolium multiflorum Lam.) fed with orchardgrass (Dactylis glomerata L.) in continuous culture. Diets were randomly assigned to fermentors in a 4 × 4 Latin square design using 7 d for adaptation and 3 d for collection. Diets were: 1) 50% orchardgrass + 50% annual ryegrass (ARG); 2) 50% orchardgrass + 50% canola (CAN); 3) 50% orchardgrass + 50% rapeseed (RAP); and 4) 50% orchardgrass + 50% turnip (TUR). Feedings (82 g DM/d) occurred four times daily throughout 4, 10-d periods at 730, 1030, 1400, and 1900 h. Methane samples were collected every 10 min using a photoacoustic gas analyzer (LumaSense Technologies, Inc.; Santa Clara, CA) during the last 3 d of the experiment. Effluent samples were collected on d 8, 9, and 10, composited by fermentor, and analyzed for VFA and pH as well as DM, OM, CP, and fiber fractions for determination of nutrient digestibility. Forage samples were analyzed for CP, NDF, ADF, minerals, and glucosinolate (GLS) concentrations. Data were analyzed using the GLIMMIX procedure of SAS. Apparent DM, OM, and NDF digestibilities and true DM and OM digestibilities were similar (P > 0.28) among diets (45.1, 63.2, 44.1, 67.1, and 87.2%, respectively). Total VFA (87.2 mol/100 mol), pH (6.47), and acetate (A: 44.6 mol/100 mol) were also not different (P > 0.20) among diets. The A:P (P = propionate) ratio was greater (P < 0.01) in ARG and CAN than RAP and TUR. Daily CH4 production was greater (P < 0.01) in ARG than all other diets (68.9 vs. 11.2 mg/d). Methane, whether expressed as g per g of OM, NDF, digestible OM, or digestible NDF fed was greatest (P < 0.01) in ARG but similar (P > 0.18) among brassica diets. A significant negative correlation was observed between total GLS and CH4 production. However, when multiple regression analysis on CH4 production was completed, neither total GLS nor individual GLS were a significant component of the model. Addition of brassicas provided similar nutrient digestibility to ARG while reducing daily CH4 production, potentially making brassicas an alternative for ARG in pasture-based ruminant diets.

    摘要:Heat stress (HS) and immune challenges negatively impact nutrient allocation and metabolism in swine, especially due to elevated heat load. In order to assess the effects of HS during Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) infection on metabolism, 9-wk old crossbred barrows were individually housed, fed ad libitum, divided into four treatments: thermo-neutral (TN), thermo-neutral PRRSV infected (TP), HS, and HS PRRSV infected (HP), and subjected to two experimental phases. Phase 1 occurred in TN conditions (22 °C) where half the animals were infected with PRRS virus (n = 12), while the other half (n = 11) remained uninfected. Phase 2 began, after 10 d with half of the uninfected (n = 6) and infected groups (n = 6) transported to heated rooms (35 °C) for 3 d of continuous heat, while the rest remained in TN conditions. Blood samples were collected prior to each phase and at trial completion before sacrifice. PPRS viral load indicated only infected animals were infected. Individual rectal temperature (Tr), respiration rates (RR), and feed intakes (FI) were determined daily. Pigs exposed to either challenge had an increased Tr, (P < 0.0001) whereas RR increased (P < 0.0001) with HS, compared to TN. ADG and BW decreased with challenges compared to TN, with the greatest loss to HP pigs. Markers of muscle degradation such as creatine kinase, creatinine, and urea nitrogen were elevated during challenges. Blood glucose levels tended to decrease in HS pigs. HS tended to decrease white blood cell (WBC) and lymphocytes and increase monocytes and eosinophils during HS. However, neutrophils were significantly increased (P < 0.01) during HP. Metabolic flexibility tended to decrease in PRRS infected pigs as well as HS pigs. Fatty acid oxidation measured by CO2 production decreased in HP pigs. Taken together, these data demonstrate the additive effects of the HP challenge compared to either PRRSV or HS alone.

    摘要:Two experiments were conducted to evaluate the effects of a high concentrate, s.c. PGF2α compared with a conventionally concentrated, i.m. PGF2α in estrus synchronization protocols for heifers. In Exp. 1, 869 Angus-based beef heifers were enrolled at 8 locations. All heifers were exposed to the 7-d CO-Synch + controlled internal drug release (CIDR) estrus synchronization protocol. On day 7 of the protocol heifers received 100 µg of GnRH i.m., and a CIDR insert for 7 d. On day 0, at CIDR removal, estrous detection patches were applied to heifers and, within location, heifers randomly received 1 of 2 PGF2α treatments: 5 mL of Lutalyse i.m. (CONTROL; n = 434) or a 2 mL of Lutalyse HighCon s.c. (HiCON; n = 435). A second GnRH injection was administered at 54 ± 2 h and heifers were fixed-time AI (TAI). Heifers were evaluated for estrous activity at TAI by determining the activation of estrous detection patches. Pregnancy rates to AI (PR/AI) were diagnosed by transrectal ultrasonography between 35 and 55 d after TAI. The percentage of heifers exhibiting estrus between day 0 and TAI did not differ (P = 0.68) between CONTROL and HiCON treatments (47 vs. 46 ± 4%, respectively). Additionally, PR/AI were similar (P = 0.65) between CONTROL and HiCON treatments (46 vs. 45 ± 3%). In Exp. 2, 190 Angus-based beef heifers were enrolled at 2 locations. Heifers were exposed to the melengestrol acetate (MGA)—PGF2α protocol where they were offered 0.5 mg MGA per day from days 1 to 14. On day 33, heifers were randomly assigned to receive CONTROL (n = 95) or HiCON (n = 95) treatment, and estrous detection aids were applied. Heifers were exposed to AI 12 h after detection of estrus. Heifers not detected in estrus at location 1 received a second PGF2α injection 6 d after the initial PGF2α injection and were placed with fertile bulls. Heifers at location 2 that did not express estrus were administered 100 µg of GnRH i.m. and exposed to TAI 96 h after the initial PGF2α injection. Transrectal ultrasonography was used to diagnose PR/AI between 51 and 57 d after the initial PGF2α injection. The percentage of heifers exhibiting estrus during the estrus detection period was similar (P = 0.40) between CONTROL and HiCON treatments (82 vs. 87 ± 4%). Furthermore, PR/AI were similar (P = 0.62) between CONTROL and HiCON treatments (60 vs. 65 ± 5%). In summary, the 2 concentrations and corresponding routes of administration of PGF2α were similar in efficacy at synchronizing estrus in beef heifers.

    摘要:Dietary fiber content and composition affect microbial composition and activity in the gut, which in turn influence energetic contribution of fermentation products to the metabolic energy supply in pigs. This may affect feed efficiency (FE) in pigs. The present study investigated the relationship between the fecal microbial composition and FE in individual growing-finishing pigs. In addition, the effects of diet composition and sex on the fecal microbiome were studied. Fecal samples were collected of 154 grower-finisher pigs (3-way crossbreeds) the day before slaughter. Pigs were either fed a diet based on corn/soybean meal (CS) or a diet based on wheat/barley/by-products (WB). Fecal microbiome was characterized by 16S ribosomal DNA sequencing, clustered by operational taxonomic unit (OTU), and results were subjected to a discriminant approach combined with principal component analysis to discriminate diets, sexes, and FE extreme groups (10 high and 10 low FE pigs for each diet by sex-combination). Pigs on different diets and males vs. females had a very distinct fecal microbiome, needing only 2 OTU for diet (P = 0.020) and 18 OTU for sex (P = 0.040) to separate the groups. The 2 most important OTU for diet, and the most important OTU for sex, were taxonomically classified as the same bacterium. In pigs fed the CS diet, there was no significant association between FE and fecal microbiota composition based on OTU (P > 0.05), but in pigs fed the WB diet differences in FE were associated with 17 OTU in males (P = 0.018) and to 7 OTU in females (P = 0.010), with 3 OTU in common for both sexes. In conclusion, our results showed a diet and sex-dependent relationship between FE and the fecal microbial composition at slaughter weight in grower-finisher pigs.

    摘要:Heat stress (HS) pigs reduce their voluntary feed intake (VFI) and ingestion of indispensable amino acids (AA). Increasing the dietary crude protein (CP) content may help to correct the reduced AA intake by HS pigs, but it may further increase their body heat load. Increasing the AA intake by adding free AA to the diet does not affect the heat load of HS pigs. Two 21-d experiments were conducted. In Exp. 1, 30 pigs (31.1 ± 1.2 kg initial body weight) were used to determine the performance depression because of HS. Treatments were: thermo neutral pigs fed a 22% CP control diet (TN-C); HS pigs fed the control diet (HS-C); HS pigs fed a 14% CP, AA supplemented diet (HS-AA). HS pigs had lower ADG and Lys utilization efficiency, and consumed 20 and 25% less Lys and Thr, respectively, than the TN-C pigs (P < 0.05). In Exp. 2 (comparative slaughter), 25 pigs (33.6 ± 0.65 kg initial body weight) were used to evaluate the effect of extra dietary AA either as protein-bound or free AA on the performance and carcass traits of HS pigs. Treatments were: control wheat-SBM-free Lys, Thr and Met diet (CON); diet with 30% more CP than CON (HSxP); diet added with free AA to contain at least 25% more of each AA than the recommended level (HSxAA). Ambient temperature (AT) ranged from 27.7 to 37.7°C, and body temperature (39.9 to 41.2°C) followed a similar daily pattern as the AT did. There was no dietary treatment effect on daily feed and NE intake (P > 0.10), but the Lys, Thr, and Met intake was higher in pigs fed the HSxP or HSxAA diets than in pigs fed the CON diet (P < 0.05). The daily weight gain (ADG) was not affected (P > 0.10) but G:F tended to be higher and the Lys utilization efficiency (ADG, g/g Lys intake) tended to be lower in HSxP pigs than in CON pigs (P < 0.10). The HSxAA pigs had higher ADG (P < 0.05), and tended to have higher weight of hot carcass and leg muscle, and the weight gain of hot carcass and leg muscle than the CON pigs (P < 0.10). The weight and daily weight gain of loin muscle was higher in the HSxAA than in the HSxP pigs (P < 0.05). Kidney weight and serum urea in HSxP pigs were higher than in CON and HSxAA pigs, but spleen weight was higher in HSxAA pigs than in CON and HSxP pigs (P < 0.05). These results confirm that HS reduces the VFI, and show that increased levels of AA either as free or protein-bound do not additionally reduce the VFI of HS pigs. These also show that extra free AA supply rather than protein-bound AA better ameliorate the reduced growth performance of HS pigs.

    摘要:Tail-end dehulling of canola meal (CM) has been shown to reduce dietary fiber and increase crude protein content in the dehulled meal. The application of this procedure also increased the total and non-phytate P content in the dehulled meal. However, it is unclear if dehulling affects P digestibility in the different fractions (i.e., the dehulled meal and the coarse fraction) and if it differs when fed to growing pigs at two different BW. Therefore, two experiments were conducted to determine the apparent (ATTD) and standardized (STTD) total tract digestibility of P in dehulled CM fed to growing pigs. Diets containing non-dehulled regular canola meal (RCM), and two fractions produced using sieve size of 355 µm: a low-fiber high-protein fraction (dehulled canola meal [DCM]) and a high-fiber low-protein fraction (coarse canola meal [CCM]) as the only source of P were fed to growing pigs at two different BW. A total of 48 pigs were used for the two experiments. In experiment 1, 24 barrows [(Yorkshire × Landrace) × Duroc] with initial BW of 24.5 ± 1.68 kg were individually housed in metabolism crates and fed the experimental diets for 10 d for total fecal collection. In experiment 2, 24 barrows with an average initial BW of 73.8 ± 4.93 kg were used; experimental diets and fecal collection procedures were the same as in experiment 1. Each experiment used six replicates per treatment. A P-free diet was used to determine basal endogenous losses of P (139.6 ± 10.7 and 150.89 ± 20.1 mg/kg of DMI for experiments 1 and 2, respectively). Data were analyzed as a completely randomized design. In experiment 1, the ATTD and STTD of P were greater (P < 0.05) for DCM (42.4% and 46.1%) than for the RCM (32.0% and 35.7%) and CCM (24.5% and 28.4%) diets. In experiment 2, the ATTD and STTD of P were greater (P < 0.05) for DCM (38.7% and 42.8%) than for the CCM diet (22.6% and 26.8%); whereas the values for RCM diet were intermediate (31.0% and 35.0%) and not different from the DCM and CCM. In conclusion, dehulling canola meal increased ATTD and STTD of P in growing pigs of different BW; however, there was no effect of BW.

    摘要:An experiment was conducted to determine the digestible energy (DE), metabolizable energy (ME), and net energy (NE) contents of canola meal (CM) and to investigate the effects of basal diet [corn diet vs. corn-soybean meal (SBM) diet] and methodology (difference method vs. regression method) on energy values of CM. Thirty-six growing barrows (20.8 ± 1.0 kg initial body weight [BW]) were individually housed in metabolism crates and randomly allotted to one of six dietary treatments to give six replicates per treatment. The six experimental diets included a corn diet, a corn-SBM diet, a corn diet with 15 or 30% of CM, and a corn-SBM diet with 15 or 30% of CM. The DE, ME, and NE of CM were determined using the corn diet or the corn-SBM diet as a basal diet. In each basal diet, two additional diets containing 15 or 30% of CM were formulated to compare the determined energy values by the difference method and estimated energy values from the regression method. Feeding level was set at 550 kcal ME/kg BW0.6 per day. Pigs were fed experimental diets for 16 d including 10 d for adaptation and 6 d for total collection of feces and urine. Pigs were then moved into indirect calorimetry chambers to determine 24 h heat production (HP) and 12 h fasting HP. The DE, ME, and NE of CM determined by the difference method were within the 95% confidence intervals estimated for the DE, ME, and NE of CM by the regression method regardless of the basal diets used, which indicates that the difference and regression methods give equivalent DE, ME, and NE of CM. However, when the goodness of fit for the linear model was compared, the r2 of the regression analysis from the corn-SBM diet (0.78) was relatively greater than that from corn diet (0.40). The estimated NE of CM by the prediction equations generated by either the corn diet or corn-SBM diets were 2,096 kcal/kg and 1,960 kcal/kg (as-fed basis), respectively, whereas those values determined by the difference method were 2,233 kcal/kg and 2,106 kcal/kg (as-fed basis), respectively. In conclusion, the NE of CM determined in the current study was, on average, 2,099 kcal/kg (as-fed basis). The difference and regression methods do not give different NE value of CM fed to growing pigs. Although the NE values of CM determined using either the corn diet or the corn-SBM diet were not different, the greater r2 of the regression analysis from the corn-SBM diet than that from the corn diet suggests that the corn-SBM diet is a more appropriate basal diet for NE determination of ingredients.

    摘要:During semen cryopreservation, spermatozoa are exposed to physical and chemical stressors that result in their functional and structural damage. Growing evidence suggests that most cryoinjuries result from oxidative stress accompanying sperm cryopreservation. Elevated amounts of reactive oxygen species (ROS) generated during cryopreservation can react with sperm macromolecules, including proteins. The goal of this study was to investigate the oxidative modifications (measured as carbonylation level changes) of carp spermatozoa proteins triggered by the cryopreservation process. Flow cytometry and computer-assisted sperm analysis were used to evaluate changes in viability, ROS level, and motility of spermatozoa. The spermatozoa proteins that were specifically carbonylated were identified and quantified by Western blotting, in conjunction with 2-dimensional electrophoresis (2D-oxyblot) and matrix-assisted laser desorption/ionization time-of-flight/time-of-flight mass spectrometry. Cryopreservation decreased spermatozoa motility (P < 0.01) and viability (P < 0.0001) and significantly increased (P < 0.0001) the number of ROS-positive cells. We identified 25 protein spots, corresponding to 19 proteins, with increases (P < 0.05) in carbonylation level due to freezing/thawing. The identified proteins are involved in motility, metabolism, calcium-ion binding, signal transduction, protein folding, and intracellular transport. The results suggest that carbonylation of flagellar proteins can result in motility disorders and may contribute to the reduced percentage of motile spermatozoa and disturbances in movement trajectory after sperm cryopreservation. Moreover, cryopreservation may contribute to impaired cellular respiration, ATP regeneration, disturbances of Ca2+ turnover, unfolding of cytoplasmic or histone proteins, disturbances of cell signaling and intracellular transport, and reduced membrane stability. Our results contribute to the knowledge concerning cryoinjury and to further development of a modified cryopreservation procedure aimed at minimizing oxidative damage of carp sperm proteins.

    摘要:To determine the effects of administration of 25 mg of PGF2α 7 d prior to the initiation of the 7-d CO-Synch + controlled internal drug release (CIDR) fixed-time AI (TAI) protocol, 985 Bos taurus beef heifers were enrolled in a completely randomized design at 9 locations from April to July of 2016. Within location, all heifers were randomly assigned to 1 of 2 treatments: 1) CONTROL (n = 496); 100 µg injection of GnRH and a CIDR insert for 7 d [day 7], administration of 25 mg of PGF2α at CIDR removal [day 0], followed by a second injection of GnRH and TAI 54 ± 2 h later; or 2) PRESYNCH (n = 489); same as CONTROL but heifers received an additional injection of 25 mg of PGF2α 7 d prior [day 14] to CIDR insertion. Estrous detection patches were applied to all heifers on day 14 and were evaluated for estrual activity on day 7. Similarly, estrus alert patches were placed on all heifers on day 0 and evaluated for estrual activity at the time of TAI. Pregnancy was diagnosed via transrectal ultrasonography between 35 and 55 d after TAI. The percentage of heifers exhibiting estrus between days 14 and 7 was greater (P < 0.001) for the PRESYNCH (70.1 ± 2.4%) than the CONTROL (41.1 ± 2.3%) treatment, whereas the percentage of heifers exhibiting estrus between day 0 and TAI was greater (P < 0.001) for the CONTROL (55.6 ± 2.4%) than the PRESYNCH (39.7 ± 2.5%) treatment. Estrus response rates differed (P < 0.001) among locations. Pregnancy rates to TAI differed (P = 0.023) among locations; however, they did not differ (P = 0.739) between CONTROL and PRESYNCH treatments (45.4 ± 2.5 vs. 43.2 ± 2.5%, respectively). Final breeding season pregnancy rates did not differ (P = 0.811) between treatments. Therefore, an injection of PGF2α 7 d prior to initiation of the 7-d CO-Synch + CIDR protocol failed to improve pregnancy rates to TAI in replacement beef heifers.

    摘要:Effects of dietary energy level and intake of corn by-product-based diets on antibody production, acute phase protein response, stress, and immunocompetency of healthy and morbid newly received growing cattle were evaluated. Four dietary treatments were formulated to supply 0.99, 1.10, 1.21, and 1.32 Mcal NEg/ kg DM and were offered at 100%, 95%, 90%, and 85% of ad libitum based on 0.99/100 treatment intake, respectively. Thirty-two pens were utilized with approximately 12 animals/pen. Four animals from each pen (32/dietary treatment) were randomly selected and used to serve as a subset to monitor immune function and acute phase proteins following a split-plot design. In addition, two animals were randomly and independently selected from each pen (16/dietary treatment) and used to measure fecal cortisol metabolite. Additionally, animals removed from the pen one (M1), two (M2), or three (M3) times and classified as morbid were bled in conjunction with a healthy control (H) removed at the same time and the serum analyzed for the same parameters. A quadratic response to time (P < 0.01) was detected for haptoglobin concentrations and for antibody titers for bovine viral diarrhea type 1 (BVD-I) and infectious bovine rhinotracheitis (IBR; P < 0.01). Haptoglobin was lowest on arrival, highest on day 14, and similar to baseline levels by day 27. Titer levels for BVD-I and IBR were lowest on arrival, higher on day 14, and significantly higher on day 27. Titers for bovine viral diarrhea type 2 (BVD-II) responded linearly (P < 0.05) with lower levels on arrival and highest levels on day 27. Haptoglobin was elevated in morbid animals compared to healthy pen mates (P < 0.05). Titer levels for BVD-I and IBR were also higher in healthy animals compared to animals pulled for morbidity (P < 0.01). Fecal cortisol was higher on arrival than on day 14 (P < 0.05). Dietary treatment had no effect on any of the parameters investigated. In summary, high-energy receiving diets based on fermentable fiber from by-products can be fed to newly received growing cattle without negative effects on antibody production toward vaccines, inflammation, or overall stress. In addition, haptoglobin concentrations and titer levels for BVD-I and IBR viruses are higher in healthy animals compared to sick animals.

    摘要:The hypothesis of this experiment was that increasing the inclusion level of distiller’s dried grains with solubles (DDGS) in the diets would decrease semen quality but have no negative effects on growth performance. Following the removal of DDGS from the diet, it was hypothesized that the ram lambs would recover and become reproductively sound, independent of treatment. To test this hypothesis, Suffolk and Hampshire ram lambs (n = 112) were allocated to 4 treatments (n = 4 pens per treatment; 7 rams per pen) in a completely randomized design. Dietary treatments were 60% corn, 25% oats, and 15% commercial lamb pellet (CON), 15% of the ration as DDGS substituted for corn (% DM basis; 15DDGS), 30% of the ration as DDGS substituted for corn (% DM basis; 30DDGS), and 45% of the ration as DDGS substituted for corn (% DM basis; 45DDGS). Lambs were fed for 112 d on their respective treatment, after which they were placed on the CON ration until day 168. Lambs were weighed on consecutive d at the beginning (days 0 and 1) and end (days 167 and 168) of the study. Scrotal circumference was measured on all lambs on days 84, 112, 140, and 168. Semen samples were collected on a subset of 64 rams (4 rams per pen) to evaluate semen quality on days 84, 112, 140, and 168. Blood samples were collected on the same subset of rams every 14 d throughout the study. A quadratic effect on BW on day 112 and overall BW (P = 0.03 and P = 0.005, respectively), ADG on day 112 and overall ADG (P = 0.02 and P = 0.02, respectively), DMI (P = 0.007) on day 112, and a cubic effect (P = 0.05) for overall G:F were observed. Overall and day 168 scrotal circumference had a quadratic (P = 0.05) response. A linear increase in spermatozoa concentration on day 168 was observed (P = 0.03) as DDGS concentration increased in the treatment diets, although rams in this stage of the study were no longer receiving DDGS. Overall, testosterone concentrations exhibited a linear decrease (P = 0.005) as DDGS increased in the diet. The linear increase (P = 0.04) on day 168 in morphologically normal sperm as DDGS increased in the diets indicated that all rams, regardless of treatment, would have passed a reproductive soundness exam. In the current study, no negative effects were observed in lamb feedlot or reproductive traits due to increasing DDGS in the diet, indicating that DDGS may be included at levels up to 45% of growing lamb diets.
