Sensitive detection of porcine circovirus 3 by dropletdigital PCR

Porcine circovirus 3 (PCV-3) is a newly emerging virus that poses apotential threat to the swine industry. We developed a sensitive assayutilizing droplet digital PCR (ddPCR) to detect PCV-3. Specificity ofthe assay was confirmed by the failure of amplification of DNA ofother relevant viruses. The detection limit for ddPCR was 1 copy/μL,10 times greater sensitivity than TaqMan real-time PCR (rtPCR). Bothmethods showed a high degree of linearity, although TaqMan rtPCRshowed less sensitivity than ddPCR for clinical detection. Ourfindings indicate that ddPCR might offer faster and improvedanalytical sensitivity for PCV-3 detection.
