龙葵氯仿提取物对荷瘤鼠肿瘤生长及脾和胸腺指数的影响

         

摘要

目的:观察龙葵氯仿提取物对荷瘤鼠肿瘤生长及脾和胸腺指数的影响。方法建立Lewis肺癌移植瘤模型,将小鼠随机分为5组,荷瘤模型组予生理盐水(0.2mL/10g)灌胃,龙葵氯仿提取物高、中、低剂量组分别按50、25、12.5mL/kg剂量的龙葵氯仿提取物灌胃,环磷酰胺组予环磷酰胺(CTX)(20mg/kg)腹腔注射(每隔2天1次),共15天。停药后24h,处死小鼠,分别剥离瘤块、胸腺、脾脏组织称重,测定各组小鼠的肿瘤生长抑制率、脾指数和胸腺指数。结果氯仿高、中、低剂量组抑瘤率分别为25.36%、38.93%、13.57%;CTX组为51.43%。环磷酰胺组及氯仿高、中、低剂量组平均瘤重分别为(1.36±0.07)g、(2.09±0.16)g、(1.71±0.11)g和(2.42±0.07)g,与荷瘤模型组(2.80±0.04)g比较,差异有统计学意义(P<0.01、P<0.05)。与荷瘤模型组比较,氯仿中剂量组的胸腺指数和脾指数显著提高(分别为130.85%、131.04%),差异有统计学意义(P<0.01),CTX组则显著降低(分别为46.24%和74.69%),差异有统计学意义(P<0.01)。结论龙葵氯仿提取物具有抑制荷瘤小鼠移植瘤生长的作用,其机制可能为通过增加荷瘤小鼠的胸腺和脾脏的重量,抑制荷瘤小鼠胸腺和脾脏萎缩,增加小鼠抗肿瘤免疫反应。%Objective To investigate the effects of chloroform extract from Solanum nlgrum L. on the growth of tumor, spleen index and thymus index of mice. Methods C57BL/6J mice were inoculated with Lewis lung carcino-ma cells and then were randomly divided into 5 groups: model group, high- , middle- and low-dosage chloroform extract groups, and cyclophosphamide group, with 8 mice in each group. Chloroform extract groups were given 50, 25, 12.5ml/kg chloroform extract of Solanum nlgrum L. by gavage, respectively. Cyclophosphamide group was given 20mg/kg of the drug by intraperitoneal injection every 3 days. Model group was given normal saline(0.2mL/10g) by gavage. The process lasted for 15d. After drug withdrawal for 24h, the mice were sacrificed and the tumor mass, thymus and spleen were taken out to be weighted for evaluation of inhibition rate, spleen index and thymus index. Results The inhibition rates were 25.36%, 38.93%, 13.57%, and 51.43% in high- , middle- , and low-dosage chloroform extract groups and cyclophosphamide group. The average tumor weights of cyclophosphamide group and high- , middle- , and low-dosage chloroform extract groups were 1.36 ±0.07g, 2.09 ±0.16g, 1.71 ±0.11g, 2.42 ±0.07g, respectively, which significantly differ from that of model group (2.80 ±0.04; P<0.01 or P<0.05). Compared with those of model group (13.58 ±1.95mg/10g and 147.40 ±13.90mg/10g), the thymus and spleen indexes of middle-dosage chloroform extract group increased 130.85% and 131.04%, respectively (P<0.01), and those of cyclophos-phamide group decreased 46.24% and 74.69%, respectively(P<0.01). Conclusion Chloroform extract from Solanumnlgrum L. may inhibit the growth of tumor in mice by enhancing the anti-tumor immune response of body through increasing thymus and spleen weights and suppressing their atrophy caused by tumor.
