外用中药对失神经支配创面VEGF表达的影响

             

摘要

目的:观察外用中药对大鼠失神经支配创面血管内皮细胞生长因子(VEGF)表达的影响。方法120只SD大鼠被随机分成4组:A组(龙血膏组)、B组(生肌膏组)、C组(失神经凡士林纱条换药组)和D组(非失神经凡士林纱条换药组)。对A组、B组、C组大鼠制造背部失神经支配的深创面模型,之后各组创面分别以龙血生肌膏、生肌愈皮膏、凡士林外用治疗,D组大鼠背部制作深创面模型,以凡士林治疗。于4、7、11、15、21d各时间点测算各组的创面愈合率,并对创面组织进行VEGF的免疫组化染色观察。结果各时间点A、B组创面愈合率均高于C、D组(P<0.05),C组低于D组(P<0.05)。A、B、D组VEGF阳性表达第11天达峰值,C组第15天达峰值。第4、7、11天时A、B组VEGF阳性表达明显高于C、D组(P<0.05),C组低于D组(P<0.05);第21天时A、B、D组低于C组(P<0.05)。结论外用中药对失神经支配创面的愈合有促进作用,可能与外用中药能增强失神经支配创面早、中期VEGF表达有关。%Objective To observe the effect of the external use of Chinese medicine on the expression of vascular endothelial growth factor(VEGF) in denervated wound healing of rat. Method 120 SD rats were randomly divided into 4 groups:A group (longxuegao group), B group(shengjigao group), C group (denervated group) and D group(normal group).A wound with denervated whole layer skin defection was established on the back of every rat in A,B,C groups,while a wound with whole layer skin defection was established on the back of every rat in D group.The wounds in A group were externally applied with longxueshengjigao,and the wounds in B group were externally applied with shengjiyupigao.The wounds of C group and D group were applied with vaseline.At different time points of 4,7,11,15,21days after modeling, the healing rate of wound were calculated.The VEGF expression of every wound was observed by immunohistochemistry stain. Results The healing rate of the A group and B group were higher than that of C group and D group(P<0.05),while the healing rate of C group was lower than that of D group(P<0.05).The peak time of C group was later than that of A group,B group and D group in VEGF expression. Observed from the volume of VEGF, A group and B group were significantly higher than the other groups in 4,7,11days(P<0.05),C group was lower than D group in 4,7,11 days(P<0.05);C group was lower than the other groups in 21 days(P<0.05). Conclusion The external use of Chinese medicine can promote wound healing of denervation,and the mechanism of promoting the wound healing may be related to enhancing VEGF expression in early and middle healing process of the wound of denervation.
