MACC1蛋白在结直肠癌患者血清中的表达及其临床意义

             

摘要

目的:探讨MACC1蛋白在结直肠癌患者血清中的表达水平及其临床意义。方法采用ELISA法检测172例结直肠癌患者及119例正常人群的血清MACC1蛋白表达情况,并分析血清中MACC1的表达水平与结直肠癌临床病理特征之间的关系。结果结直肠癌患者血清中的MACC1的表达水平显著高于正常人的表达水平(P<0.05)。结直肠癌患者血清中的MACC1表达水平与性别(P=0.004)、CA19-9表达情况(P=0.039)、淋巴结转移情况(P<0.001)、远处转移情况(P=0.014)、神经侵犯情况(P=0.019)、癌结节(P=0.014)及TNM分期(P<0.001)等密切相关,而与年龄、肿瘤家族史、肿瘤位置、浸润深度、脉管瘤栓、病理分型及分化程度无明显相关性(P>0.05)。此外, MACC1高表达组的结直肠癌患者总生存率(OS)明显低于低表达组,两者差异具有统计学意义(χ2=9.466,P=0.002)。结论 MACC1在结直肠癌患者血清中表达升高,并且其表达量与结直肠癌的恶性演变及临床预后密切相关,有望成为结直肠癌诊断及预后的血清学指标。%Objective In this study,the expression of serum MACC1 protein was investigated in patients with colorectal cancer,and the clinical relevance was explored. Methods We retrospectively analyzed 172 patients with colorectal cancer in our center during the period of Aug 2011 through Aug 2012. 119 healthy serums were used as control. Serum was isolated by Centrifugation and the expression of MACC1 protein was tested by ELISA. Relationship between the expression level of MACC1 and clinical characteristics of colorectal cancer was analyzed by SPSS 18.0 analysis software. Results The expression of serum MACC1 protein in colorectal cancer patients was significantly higher than that in normal controls (P<0.05). Then the analysis was done in patients divided by risk stratification. In our research,we found that in colorectal cancer patients,serum MACC1 protein expression had a closely relationship with gender (P=0.004),CA19-9 level (P=0.039),lymph node metastasis (P<0.001),distant metastasis (P=0.014),neural invasion (P=0.019),cancer nodules(P=0.014) and TNM stage (P<0.001). But there was no significant correlation with patient age,family tumor history,tumor location,depth of tumor invasion,vascular tumor thrombus,and pathological type (P>0.05). In addition,overall survival rate(OS) in patients with high expression of MACC1 was significantly lower than that in the low expression group (χ2=9.466,P=0.002). Conclusion The expression of serum MACC1 protein was increased in patients with colorectal cancer. And the expression level was closely related to the malignant evolution and clinical prognosis in colorectal cancer. The serum MACC1 protein may be a good serological marker for diagnosis and prognosis of colorectal cancer.
