芪连扶正胶囊对 TGF-β介导 EMT 作用的影响

摘要

目的：观察芪连扶正胶囊含药血清对 TGF-β及 EMT 相关分子在肺癌 A549细胞中表达的影响，探讨相关机制。方法：体外培养肺癌 A549细胞，制备芪连扶正胶囊含药血清，分组干预，采用 RT-PCR 检测 TGF-β1mRNA 表达，IC 检测 E-cad 与 N-cad 表达。结果：对照组、芪连扶正低剂量、中剂量、高剂量组、化疗组及联合组的 TGF-β1 mRNA 相对表达量依次为（31.11±1.58）％、（29.53±1.50）％、（27.28±1.75）％、（25.23±1.38）％、（23.03±1.59）％、（20.72±1.38）％；各组E-cad、N-cad 均有不同程度阳性表达，除芪连扶正胶囊低剂量组外，各药组 E-cad 阳性率均高于对照组、N-cad 则相反（P ＜0.05或 P ＜0.01）；与化疗组相比，联合组及芪连扶正胶囊各组亦有统计学意义（P ＜0.05或 P ＜0.01）。结论：芪连扶正胶囊可通过逆转 TGF-β介导的 EMT 作用防止肿瘤转移。%Objective:To observe the effect of Qilian Fuzheng Capsule on expression of EMT(epithelial mesenchymal transition) and TGF-βin lung cancer A549 cells,and study its possible mechanism.Methods:A549 cells were cultured in vitro and group in-tervened,RT-PCR to detected the expression of TGF-β1mRNA,using Immunohistochemical to detected the expression of E-cad and N-cad.Results:the TGF-β1 mRNA relative expression of the control group,Qilian Fuzheng Capsule low,medium and high dose groups,chemotherapy group and combined group were (31.11 ±1.58)%、(29.53 ±1.50)%、(27.28 ±1.75)%、(25.23 ±1.38)%、(23.03 ±1.59)%、(20.72 ±1.38)% respectively.Except the low-dose group,the expression of E-cad and N-cad in other drug groups were all significantly different compared with the control group;the combined group and all Qilian Fuzheng Capsule groups were significantly different compared with the chemotherapy group.Conclusion:Qilian Fuzheng Capsule can reverse the TGF-β-mediated EMT therefore play an anti-tumor effect.
