To investigate feasibility of differentiation of mesenchymal stem cells (MSCs) into cardiomyoctes.To establish a new therapeutic way for heart myocardial infarction.Methods MSCs,which were separated from bone marrow,induced by 5-azacytidine for 24 hours after being cultured and proliferated.The expression of alpha-sarcomeric actin was observed after additional 14 days and 21 days,respectively.The cultured MSCs were transplanted into the myocardial infarction site and results were observed by histological methods.Ultrasound was also used to detect the ventricular wall movement and heart function.Results The expression of alpha-sarcomeric actin was observed in part of the MSCs induced by 5-azacytidine and cultured for 21 days.Spontaneous beating of the cells was also witnessed.4 weeks later,some MSCs that were transplanted into the acute myocardial infarction site had been differentiated into cardiomyocytes with typical sarcomeres and postive expression of alpha-sarcomeric actin.Conclusion MSCs can be differentiated into cardiomyocytes.7 refs,4 figs,1 tab.
