两种乳酸菌发酵液对人胃癌细胞株BGC823体外增殖及小鼠免疫功能的影响

         

摘要

Objective To explore the effect of lactobacillus (LAB) fermentation broth on the in vitro proliferation of human gastric carcinoma cell line BGC823 and the immunologic function of mice,which will provide the basis for the prevention and treatment of tumors.Methods Lactobacillus bulgaricus,Streptococcus thermophilus were cultivated and fermented.MTT was used to determine the proliferative activity of human gastric carcinoma cell line BGC823 treated by two fermentation broths,the PBS control group was set.The mice were treated by intragastric administration of different fermentation broths.The change of the foot pad thickness was carried out to evaluate delayed type hypersensitivity (DTH),spleen weight and spleen index were detected,and the specific antibody formation ability was detected by using complement hemolytic assay; the normal saline group was set.Results Compared with the control group,cell proliferative activity of BGC823 cells was slightly decreased after the treatment of two kinds of fermentation broths and mixture of them (all P >0.05),while the mouse foot pad thickness difference,spleen weight,spleen index and antibody hemolysin level were significantly increased (all P < 0.05).Conclusion The inhibitory effects of Lactobacillus bulgaricus and Streptococcus thermophilus fermentation broths on the proliferation of gastric cancer cells are not obvious,but they can enhance immunologic function in mice.All the above can be expected to provide new ideas for tumor prevention and treatment.%目的 观察乳酸菌(LAB)发酵液对人胃癌细胞株BGC823增殖及小鼠免疫功能的影响,为肿瘤的防治提供依据.方法 培养保加利亚乳杆菌、嗜热链球菌并分别制备发酵液,采用四氮唑蓝(MTT)比色法检测其对人胃癌细胞株BGC823增殖活性的影响,设PBS对照;用发酵液对小鼠连续灌胃给药,通过足垫厚度增加法观察绵羊红细胞诱导的小鼠迟发型超敏反应(DTH)、检测脾脏质量和脾脏指数、运用补体溶血实验检测特异性抗体形成能力,设生理盐水对照.结果 与对照相比,两种发酵液及其混合液处理后BGC823细胞的增殖活性略有降低(P均>0.05),小鼠足垫厚度差值、脾脏质量、脾脏指数、抗体溶血素水平均显著增高(P均<0.05),以混合发酵液处理者为著.结论 保加利亚乳杆菌、嗜热链球菌发酵液对胃癌细胞增殖的直接抑制效果不明显,但对小鼠的细胞免疫与体液免疫均具有增强作用;本研究为肿瘤防治提供新了思路.
