喉癌组织中Ccbe1、VEGF-D的表达变化及意义

         

摘要

目的 观察喉癌组织中胶质和钙结合EGF区域1(Ccbe1)、血管内皮生长因子D(VEGF-D)的表达变化,并探讨其意义.方法 采用免疫组化法检测45例喉癌组织(喉癌组)及20例因声带息肉手术切除的喉良性组织(良性组)中的Ccbe1、VEGF-D,分析两指标间及与喉癌临床病理参数、染色计数淋巴管密度(LVD)、CD34染色计数微血管密度(MVD)的相关性,观察喉癌组Ccbe1表达阳性及阴性者生存时间.结果 喉癌组Ccbe1、VEGF-D的阳性表达率分别为51.1%、60%,对照组分别为20%、25%,两组比较P均<0.05.Ccbe1表达与喉癌TNM分期、淋巴结转移相关(P均<0.05).喉癌组织中Ccbe1与VEGF-D表达呈正相关(r=0.381,P<0.05).Ccbe1和VEGFD表达均与MVD、LVD密切相关(P均<0.05).Ccbe1阳性者生存时间为(43.94 ±5.47)个月,阴性者生存时间为(61.09 ±4.75)个月,两者比较P<0.05;Ccbe1阳性表达与生存曲线呈负相关(P<0.05).结论 Ccbe1能够通过上调VEGF-D表达促进喉癌淋巴管生成和血管生成,检测Ccbe1表达可成为判断喉癌预后新的生物学指标.%Objective To observe the expression changes of collagen and calcium binding EGF domains 1 (Ccbe1) and vascular endothelial growth factor D (VEGF-D) in patients with laryngeal cancer,and to investigate their significance.Methods Forty-five specimens of the laryngeal cancer tissue (group A) and 20 specimens of benign laryngeal tissue (group B) were studied.The protein expression of Ccbe1 and VEGF-D were detected by immunohistochemistry,LVD was evaluated by staining,MVD was evaluated by immunohistochemistry with CD34; the correlation analysis was made between two factors and clinical parameters,LVD,MVD.The survival time of the Ccbe1 positive and negative groups in laryngeal carcinoma was observed,Results The positive protein expression of Ccbe1 and VEGF-D were 51.1% and 60% in tumor tissue,and those in the normal tissue were 20% and 25%,comparison of two group were both P <0.05.The expression of Ccbe1 was significantly correlated with TNM stage and lymphatic metastasis (P < 0.05).The expression of Ccbe-1 was significantly positive correlated with the expression of VEGF-D (r =0.381,P < 0.05).The expression of Ccbel and VEGF-D was significantly correlated with MVD and LVD (P < 0.05).The survival time in the Ccbe1 positive group was 43.94 ± 5.47 months,the survival time in the Ccbe1 negative group was 61.09 ± 4.75 months (P < 0.05).The positive expression of Ccbe1 was negatively correlated with the survival curve (P < 0.05).Conclusions Ccbe1 promote lymphangiogenesis and angiogenesis in laryngeal cancer by up-regulating the expression of VEGF-D.Detection of Ccbe1 protein may be helpful for judging prognosis in laryngeal cancer.
