东亚钳蝎毒素对人结肠癌细胞 Caco －2的增殖抑制作用研究

摘要

为研究东亚钳蝎毒素对人结肠癌细胞 Caco －2增殖的影响，以不同浓度的东亚钳蝎毒素（10、20、40μg／mL）干预体外培养的 Caco －2细胞，分别于24、48 h 后，用四甲基偶氮唑盐（MTT）比色法，观察毒素对Caco －2细胞的增殖抑制作用，运用淋巴细胞转化试验和乳酸脱氢酶（LDH）释放试验检测蝎毒素对 Caco －2细胞的作用途径。结果表明：东亚钳蝎毒素不仅能抑制 Caco －2细胞的增殖而且能促进淋巴细胞转化，且毒素对 Caco －2细胞增殖的抑制作用具有浓度和时间的依赖关系，随浓度加大和时间延长，对 Caco －2细胞增殖的抑制作用增强。%The effects of Buthus martensii Karsch venom on proliferation of human colon adenocarcinoma cell line Caco -2 were investigated in this study.The test used different concentrations (10,20,40 μg/mL) of Buthus martensii Karsch venom to interfere the Caco -2 cells cultured in vitro for 24 and 48 hours,then the proliferation inhibition effects was observed by means of MTT assay.The mechanism of venom on Caco -2 cells was measured by lymphocyte transformation and lactate dehydrogenase release assays.The data indicated that Buthus martensii Karsch venom could inhibit the Caco -2 cell proliferation and promote the lymphocyte transformation.Besides,the inhibition effects were closely related to the treatment time and venom concentra-tion,which enhanced with the extension of treatment time and increase of venom concentration.
