木犀草素预处理对心肌缺血再灌注损伤大鼠炎症反应及细胞凋亡的影响

摘要

Obj ective:To detective the effects of luteolin pretreatment on inflammatory reaction and apoptosis of myocardium of ischemia-reperfusion(I/R)inj ury in rats.Method:Male wistar rats were randomly allocated into three groups:sham group (group A),I/R group (group B),luteolin group (Group C),with 10 rats in each group,4 days be-fore operation were inj ected via the tail vein giving corresponding treatment.The rats model of myocardial ischemia reper-fusion (MI/R)injury was induced by a transient vessel occlusion for 30 min and reperfusion for 2h.The contents of,IL-6 and TNF-were detected by ELISA,and apoptosis index of myocardial cells was detected by TUNEL method.Resuls Compared to group A,group B,myocardial apoptosis index,serum TNF-αand IL-6 levels increased significantly (P<0. 01).Results:Compared to group B,serum TNF-αand IL-6 levels and myocardial apoptosis index in group C were signifi-cantly lower (P<0.01 or 0.05).Conclusion:Luteolin pretreatment have protective effects on myocardial ischemia reper-fusion injury in rats,the mechanism of it may be through down-regulated TNF-αand IL-10 expression to reduce inflam-mation and inhibit myocardium apoptosis.%目的：：研究木犀草素预处理对大鼠心肌缺血再灌注损伤中细胞凋亡和炎症反应的影响。方法：以30只健康雄性 Wistar 大鼠为研究对象,随机分为假手术组(A 组),缺血再灌注组(B组),木犀草素组(C组),每组10只,术前4d分别经尾静脉注射给予相应处理。阻断冠脉左前降支,构建心肌缺血模型,缺血30min 后再灌注2h。用 ELISA 法检测大鼠血清 TNF-α, IL-6的含量,采用TUNEL法检测心肌细胞的凋亡指数。结果：与 A 组相比,B组心肌细胞凋亡指数、血清TNF-α及IL-6含量明显上升(P<0.01)；与B组比较,C组的心肌细胞凋亡指数,血清IL-6和TNF-α的含量则显著降低)(P<0.05或0.01)。结论：木犀草素预处理可以抑制心肌缺血再灌注导致的心肌细胞凋亡,其机制可能与减少促炎细胞因子 TNF-α及IL-6的产生,进而减轻炎症反应有关。
