风水肾炎汤对IgA肾病大鼠模型治疗作用及血清Scr、BUN、IL-21表达的研究

摘要

目的:研究自拟风水肾炎汤治疗IgA 肾病大鼠模型效果及对血清Scr、BUN、IL-21、IL-4、T N F-α表达水平的影响.方法:购进100只实验用健康大鼠,随机均分为空白组、模型组、阳性药物组及治疗组四组.空白组正常喂养,其它三组制备为IgA 肾病大鼠模型后模型组不予任何治疗、阳性药物组给予安慰剂、治疗组给予自拟风水肾炎汤治疗;治疗或喂养1个月后对比各组大鼠24 h尿蛋白定量、血清尿素氮(BUN)、肌酐(Scr)、肿瘤坏死因子(TNF-α)、白细胞介素-4(IL-4)、白细胞介素-21(IL-21)含量.结果:各组间24 h尿蛋白、Scr、BU N均具有统计学差异(P<0.05);各组间T N F-α、IL-4、IL-21均具有统计学差异(P<0.05).结论:风水肾炎汤对于Ig A肾病模型大鼠具有明确治疗作用,能够降低其尿液中蛋白含量以及其血清中肌酐、尿素氮水平,通过降低其血清中IL-21调控免疫反应实现保护肾功能的作用.%Objective:To study the effect of self-made Fengshui Shenyan decoction on IgA nephropathy rats and the effect on the expression of serum Scr,BUN,IL-21,IL-4 and TNF-alpha.Methods:The purchase of 100 experimental healthy rats were randomly divided into control group,model group,positive drug group and treatment group,control group of normal feeding,the other three groups were prepared for IgA nephropathy rats model group without any treatment,the positive drug group were given placebo,treatment group was treated by Fengshui Sheny-an decoction in the treatment of nephritis treatment;or after 1 months compared with the rats of 24 h urinary pro-tein,serum urea nitrogen(BUN),creatinine(Scr),tumor necrosis factor alpha(TNF-alpha)and interleukin-4 (IL-4),interleukin-21(IL-21)content.Results:The 24 h urine protein,Scr and BUN were statistically different in each group(P<0.05),and TNF-alpha,IL-4 and IL-21 in each group were statistically different(P<0.05).Con-clusion:Fengshui Shenyan decoction has a definite therapeutic effect on IgA nephropathy rats.It can reduce the con-tent of protein in urine and the level of creatinine and urea nitrogen in serum.It can protect renal function by decrea-sing its IL-21 in serum and regulating immune response.
