蒙药耧斗菜不同产地木兰花碱含量的比较研究

摘要

Objective To analyse the content of magnoflorine of the Mongolian medicine Aquilegia from different origins,and establish magnoflorine conent determination method and quality evaluation standard.Methods HPLC was carried out under the following conditions,the mobile phase was acetonitrile-0.05 mol/L potassium dihydrogen phosphate (12:88) (add 100 mL per 0.2 mL triethylamine,pH 4.0),the detection wavelength 286 nm,the flow rate 1.0 mL/min and the column temperature 22 ℃.Results The linear range of magnoflorine was 20.95～419.00 μg/mL (r=0.999 5,n=6),the average recovery was 101.2％,RSD was 1.240％,and the content of magnoflorine of the Aquilegia from different origins ranged from 0.07 to 0.48％.Among of them,The highest concentrations was from Qingzhou Shandong and the lowest was from Daqingshan JingErgou Hohhot.Conclusion This method was accurate and reliable,which could be used for quality control.%目的 对蒙药耧斗菜不同产地木兰花碱的含量进行比较研究,建立蒙药耧斗菜中木兰花碱的含量测定方法与质量评价标准.方法 采用高效液相色谱法,流动相为乙腈-0.05 mol/L磷酸二氢钾(12:88)(每100 mL中加0.2 mL三乙胺,再以磷酸调pH值为4.0)；检测波长为286 nm;流速为1.0 mL/min；柱温为22℃.结果 木兰花碱在20.95-419.00μg/mL范围内与峰面积积分值呈良好的线性关系(r=0.999 5,n=6),平均加样回收率为101.2％,RSD为1.240％,不同产地耧斗菜中木兰花碱含量在0.07-0.48％之间,其中以山东青州产地含量最高,以呼市大青山井儿沟产的含量最低.结论 该方法精密度、重复性、稳定性良好,方法准确、可靠,对蒙药耧斗菜药材的质量控制与质量评价提供实验依据.
