冰冻切片技术制作药用植物组织永久切片的研究

摘要

目的 本实验建立了冰冻切片技术制作药用植物组织永久切片的实验方法.方法 将材料用FAA固定液固定,经过包埋后,利用冰冻切片机在-18℃-20℃温度下切片,切片经乙醇梯度脱水、番红-固绿染色、二甲苯透明后,用加拿大树胶封片即得.结果 应用该方法制作的植物组织永久切片,结构完整而清晰.结论 冰冻切片技术是制作药用植物组织永久切片简便有效的方法,应用前景广阔.%Objective To establish the methods of making permanent sections of pharmaceutical botany tissues by cryo-sectioning technique. Method The procedure consisted of several steps as follows: The materials were embedded after fixed by FAA fixation liquid, then frozen and sliced by cryostat microtome betweent -18℃and-20℃ . Result Sections were dehydrated in ascending series of ethanol, dyed with safranine and fast green , cleared in xylene and sealed with canada balsam. Structure of the section was intact and clear. Conclusion Cryo-section technology was a simple and efficient method to make permanent sections of plant tissues, it has broad prospect of application.
