耐盐产氢菌C.beijerinckii n232的分离及产氢能力研究

         

摘要

从秦皇岛海滨泥沙样品中分离获得一株具有耐盐性的产氢细菌,编号为n232.通过生理生化鉴定和16S rDNA系统发育分析,确定该菌株为拜氏梭菌的新菌株,命名为Clostridium beijerinckii n232(KU750806).菌株n232发酵产氢的最适碳源为葡萄糖,葡萄糖的最适初始浓度为10g/L;最适初始pH为7.0;蛋白胨与酵母粉的最佳投加量分别为3g/L和2g/L;投加NaC1可以促进细胞生长,提高比产氢率.使用优化的培养基,发酵24h,菌株n232获得的最大细胞干重为1.13 g/L,最大比产氢率为2.14 mol/mol.耐盐发酵产氢细菌的筛选为高盐度有机废水资源化提供了新的思路.%A hydrogen-producing strain with NaCl tolerance capacity numbered n232 was isolated from Qinhuangdao coast sediment sample.It was identified belonging to Clostridium beijerinckii based on 16S rDNA sequencing and physiological-biochemical analysis,and named as Clostridium beijerinckii n232 (KU750806).The experiments results indicated that glucose was the most suitable carbon source,the optimum initial concentration of glucose was 10 g/L,the favorite initial pH was 7.0,the most suitable concentration of peptone and yeast powder were 3 g/L and 2 g/L,and adding suitable concentration NaCl had a promoting effect on cell growth and hydrogen yield of strain n232.The maximum dry cell weight of 1.13 g/L and hydrogen yield of 2.14 mol/mol were obtained with the optimal fermentative culture medium after 24 h fermentation.The isolation of salt tolerance hydrogen producing bacteria provides a new idea for the high salinity organic wastewater resources.
