以多浆旱生植物霸王(Zygophyllum xanthoxylum)幼苗根系为材料,采用RT-PCR方法获得了其质膜Na+/H+逆向转运蛋白基因ZxSOS1的片段,以中间载体pHANNIBAL和植物表达载体PART27为基础,采用酶切连接的方法构建了CaMV 35S启动子驱动的含ZxSOS1基因片段反向重复序列的RNAi植物表达载体PARS,为利用RNAi技术深入研究ZxSOS1在霸王体内Na+转运中的功能及其在霸王适应干旱生境中的作用机制奠定基础。%Total RNA was extracted from roots of succulent xerophyte Zygophyllum xanthoxylum seedlings,and the fragment of the plasma membrane Na+/H+ antiporter gene ZxSOS1 was amplified by RT-PCR.Then the RNAi plant expression vector PARS with inverted repeats of ZxSOS1 driven by the promoter CaMV 35S was constructed based on the intermediate vector pHANNIBAL and the plant expression vector PART27 by restriction enzyme digestion and ligation.The work might be a good basis for further investigation of the roles of ZxSOS1 in the transportation of Na+ and the adaption to arid environment in Z.xanthoxylum by RNAi technique.
