神经母细胞瘤原位荷瘤与转移瘤裸鼠模型建立的实验研究

摘要

Objective We use the established QDDQ cell lines to establish the situ and metastasis tumor-bearing nude mice model,to detect neuroblastoma metastatic characteristics. Methods We used fresh neuroblastoma tumor tissue, Use of enzyme digestion method,to establish cell lines in vitro with 0. 5 ml single cell suspension,the concentration is 3-7×107/ml,Inoculated into nude mice,to establish nude mouse model. Results We get the neuroblastoma in situ and metastatic tumor-bearing nude mice model successfully. A total of 30 experimental nude mice, Tumorigenicity of 25, Tumorigenicity of rate of 83. 3% . Conclusions The establishment of animal model with neuroblastoma cells to provide us a good research platform for the further study of the transfer characteristics, molecular biological characteristics.%目的 用自建的细胞系QDDQ建立起原位荷瘤与转移瘤裸鼠模型,探讨神经母细胞瘤(Neuroblastoma,NB)的转移特性.方法 取NB新鲜瘤组织,利用酶消化法,建立起体外细胞系,以(3～7)×107/ml浓度的单细胞悬液0.5 ml接种于裸鼠,建立荷瘤鼠模型[1].结果 NB原位荷瘤与转移瘤裸鼠模型成功建立,实验鼠共30只,致瘤25只,致瘤率为83.3%.10只原位荷瘤,15只转移荷瘤.结论 建立NB细胞动物模型,对于进一步研究其转移特性、分子生物学特征,提供了良好的平台.
