以剑麻H.11648茎尖愈伤组织为材料,采用化学诱变剂EMS(甲基磺酸乙酯)诱变处理,设置6个浓度梯度(0%、0.2%、0.4%、0.6%、0.8%和1.0%),5个处理时间(2h,4h,6h,8h和16 h)共30个处理组合,结果表明,用0.8%的EMS处理6h和1.0%的EMS处理4h,愈伤组织死亡率分别为52.22±1.11%和56.67±1.67%,分化系数分别为7.63±1.10和7.13±2.79,符合“半致死剂量”标准,可以作为筛选剑麻突变体浓度组合.%Chemical mutagenesis EMS was used to induce callus derived from the shoot tips of Agave hybrid cv No.11648.Thirty experimental treatments were designed based on two influential factors which were six concentration levels (0 % 、0.2 % 、0.4 % 、0.6 % 、0.8 % and 1.0 %),and five time points (2 h,4 h,6 h,8 h and 16 h).The results showed that the combinations of 0.8 % EMS and 6 h,1.0 % EMS and 4 h could be used to screen the sisal mutants.The lethal rates of callus were 52.22 ± 1.11% and 56.67 ± 1.67 %,the differentiation coefficients were 7.63 ± 1.10 and 7.13 ± 2.79,respectively.
