高效液相色谱法测定头孢呋辛酯中高分子聚合物

             

摘要

目的:建立HPLC法测定头孢呋辛酯中高分子聚合物的方法.方法:采用聚苯乙烯-二乙烯苯共聚物(分子量使用范围< 10000)为填充剂的色谱柱;0.030 mol·L-1 LiBr溶液为流动相;流速为0.33 mL·min-1;检测波长为280 nm;进样量为20μL.结果:头孢呋辛酯在0.104~0.304 mg·mL-1范围内,溶液浓度与聚合物峰面积呈良好的线性关系,线性回归方程为Y=353576X-3182.2(r=0.9998),检测限为0.068 ng.结论:本方法简单、快速、灵敏度高、重现性好,克服了传统聚合物检测方法分析时间长,无法检测脂溶性样品的缺点.可以用于测定头孢呋辛酯、头孢呋辛酯制剂及其它脂溶性抗生素中高分子聚合物.%Objective: To establish a method for determining the polymers in cefuroxime axetil by HPLC. Methods! The chromatographie column was packed by the copnlymers of polystyrene and di vinyl— benzene (the molecular weight was less than 10000); the mobile phase was composed of a 0.030 mol ·L-1 LiBr solution; the flow rate was at 0.33 mL·min-1; the detection wavelength was at 280 ran; the injection volume was 20 μL. Results: The calibration curve was linear in the concentration range of 0.104~0.304 mg·rnL-1 (r=0.9998), the detectabiltty was 0.068 ng. Conclusion: The method is simple, quick, sensitive and reproducible, ll can be used to determine the polymers in cefuroxime axetil, cefuroxime axetil preparations and other liposolubile antibiotics.
