胃癌组织中胸腺嘧啶合成酶和双氢嘧啶脱氢酶的表达及临床意义

         

摘要

目的 了解胸腺嘧啶合成酶(TS)和双氢嘧啶脱氢酶(DPD)mRNA在胃癌组织及癌旁非肿瘤组织中的表达情况以及与临床病理学特征的关系.方法 采用半定量反转录聚合酶链式反应(RT-PCR)方法测定50例胃癌患者TS、DPD mRNA在胃癌及癌旁非肿瘤组织的表达.结果 胃癌组织中TS mRNA水平明显高于非肿瘤组织中的水平(P=0.001),DPD mRNA水平略高于非肿瘤组织但无明显差异(P=0.107).肿瘤和非肿瘤组织中TS mRNA水平(P=0.382)、DPD mRNA水平之间(P=0.227)以及肿瘤组织中TS和DPD mRNA水平(P=0.412)、非肿瘤组织TS和DPD mRNA水平之间(P=0.412)无显著相关性.对比临床病理学特征与TS mRNA水平,Ⅲb期与Ⅳ期的水平无明显的差异(P=0.106),但却高于Ⅰa、Ⅰb、Ⅱ、Ⅲb期水平(P=0.045);在分化程度上未分化型中TS mRNA水平要高于已分化的细胞类型(P=0.017),而DPD mRNA与临床病理学特征无显著相关性.结论 胃癌组织中TS mRNA的水平高于非肿瘤组织中的水平;TS mRNA水平与临床分期有关,Ⅲb期水平高于Ⅰa、Ⅰb、Ⅱ、Ⅲa期的水平,而与Ⅵ期水平无明显差异;同时TS mRNA水平与肿瘤细胞的组织学类型有关.%Objective To investigate the correlation between TS and DPD mRNA level in tumoral and non-tumoral egastric mucosa. Methods The expressions of TS and DPD mRNA in tumoral tissue and adjacent non-tumoral mucosa of 50 patients with gastric cancer were measured by semi-quantiative reverse transcription-polymerase chain reaction (RTPCR). Results The TS mRNA level in tumor tissue was significantly higher than non-tumoral tissue (P =0. 00), and the DPD mRNA levels was higher than non-tumoral tissue, but without statistical significance (P =0. 107). There was not correlation between both the TS mRNA and DPD mRNA levels in tumoral and non-tumoral tissue (P=0. 382); also,there was not correlation between TS and DPD mRNA in tumoral ( P = 0. 412 ) and in non-tumoral ( P = 0. 412, P > 0. 05) tissues. The TS mRNA levels were relate to stage and histological type, for which no significant differene existed between stage Ⅲ b and Ⅳ (P = 0. 106 ), but it was significantly higher in stage Ⅲ b than the other (P = 0. 045 ); the TS mRNA levels in undifferentiated type is higher than that differentiated type (P =0. 017). No significant correlation was observed between tumoral DPD mRNA leves and any clinicopathologic characteristics. Conclusion The TS mRNA levels is higher in tumoral tissue of gastric cancer than that in non-tumoral tissue; The TS mRNA levels relate to clinical stage,which in stage Ⅲb is higher than in stage Ⅰa, Ⅰb, Ⅱ, and Ⅲa, but no significant difference between stage Ⅲb and Ⅳ. The TS mRNA levels relate to histological type.
