香水百合脱毒技术研究

         

摘要

An virus-free treatment was made for the robina seedball ofL.orientalhybridsby the methods of micro tip cul-ture, heat treatment and detoxification, and chemical agent,and theL.orientalhybrids treated was cultured, and the seedlings of the treatedL.orientalhybrids were detected by enzyme linked immunosorbent assay. The results showed that theL.orientalhybrids was treated with high temperature at 32 ℃for 4 weeks, the survival rate and the cucumber mosaic virus-free effect of theL.orientalhybridsrobina reached the best through following two combinations of treatments: 0.3 ~ 0.6 mm shoot tip culture and 0.3 ~ 0.6 mm shoot tip culture on ribavirin with a concentration of 15 mg/L.%通过微茎尖培养脱毒方法、热处理脱毒方法和化学药剂脱毒等方法对香水百合罗宾娜种球进行脱毒处理,并将处理百合进行培养,通过酶联免疫吸附法对处理百合继带苗进行检测.结果表明:当香水百合用32 ℃的高温处理4周后,切取0.3~0.6 mm微茎尖培养的处理、切取0.3~0.6 mm微茎尖培养在含有浓度为15 mg/L病毒唑上的处理,这两种组合法香水百合罗宾娜存活率和黄瓜花叶病毒脱毒效果达到了最佳.
