马胎盘提取物对淋巴细胞的免疫调节作用

摘要

采用MTT法测定不同浓度马胎盘提取物(EPE)对正常、ConA和LPS诱导的小鼠脾淋巴细胞增殖活性以及NK细胞杀伤活性的影响；并用氨基酸自动分析仪检测马胎盘提取物中氨基酸的含量.MTT实验结果表明:马胎盘提取物(187.5 ～750 μg/mL)明显增强正常淋巴细胞的活性；23.44 ～ 1500 μg/mL明显抑制ConA诱导的T淋巴细胞增殖活性,750～1500 μg/mL明显抑制LPS诱导的B淋巴细胞增殖活性,其对ConA诱导的T淋巴细胞增殖的抑制作用强于对LPS诱导的B淋巴细胞增殖的抑制作用；93.75 ～500μg/mL明显增强NK细胞对PC-3癌细胞的杀伤活性；氨基酸含量分析表明:其含有丰富的氨基酸,其中谷氨酸(7.53％)、天门冬氨酸(5.64％)和亮氨酸(5.54％)含量较高.因此马胎盘提取物对淋巴细胞可能具有一定的免疫调节作用.%The effects of different concentrations of the equine placental extract on the normal lymphocytes and the proliferation of lymphocytes stimulated by ConA and LPS and the cytotoxicity of NK cells against PC-3 cells were measured by MTT assay.The MTT assay results indicated that suitable concentrations(187.5-750 μg/mL) of equine placental extract can signally enhance the viability of normal lymphocytes.The proliferation of lymphocytes stimulated by ConA and LPS were notably suppressed by certain concentration of equine placental extract,particularly suppressed the proliferation of T lymphocytes stimulated by ConA.Equine placental extract (93.75-1500 μg/mL) can also enhance the cytotoxicity of NK cells against PC-3 cells.Content analysis of amino acid indicated that equine placental extract contained abundant amino acids.Equine placental extract could possess immunoregulation activity on lymphocytes.
