目的 研究中医调护干预创伤后压力心理障碍症(PTSD)孕鼠对子鼠生长发育、行为的宏观表征影响. 方法 按照随机数字表法将12只受孕SD母鼠分为空白对照组、PTSD组、情志中药组. 采用单一长期应激(SPS)法造成PTSD模型;情志中药组造模后给予金匮肾气丸混合饲料补肾,同时放在优裕环境鼠笼构造情志护理;重复观测各组存活子鼠的体质量、身长、尾长、旷场试验(OFT)表现及糖水消耗情况. 结果 PTSD组子鼠体质量增长最慢,而中药情志组体质量增长较快,且优于空白对照组(P<0.05).PTSD 组旷场试验得分低于空白对照组和情志中药组(P均<0.05), 而空白对照组与中药情志组相比差异无统计学意义. PTSD组糖水消耗量小于空白对照组和情志中药组(P均<0.05),而空白对照组与中药情志组相比差异无统计学意义. 结论 母鼠孕期PTSD可以遗传到子代,延缓子代的生长发育;金匮肾气丸联合优裕环境情志护理可纠正母鼠PTSD导致的子鼠发育迟缓,且可促进生长发育,为进一步揭示PTSD胎损的遗传和调护机制奠定了基础.%Objective It is to find out the influence of TCM nursing combined with tonifying kidney Chinese medicine on behavioral genetics of maternal SD rats with posttraumatic stress disorder (PTSD) and their offspring with fetus injury.Methods 12 pregnant rats were randomly divided into 3groups : blank control group, PTSD group, and emotion-medicine group. PTSD models were established by single prolonged stress (SPS) .After the models were established, the rats in emotion-medi-cine group were given Jin Kui Shenqi Wan mixed into conventional feed to supple their kidney and given emotional nursing through enriched environmental cage.Repeat measurements included body weight, body length, tail length, openfield test (OFT) performance, and saccharose water consumption were performed on.Resutl s The body weight increase of the offsprings in PTSD group were slowest, and the increase was faster in emotion-medicine group which was better than that in the control group (P<0.05).In OFT, PTSD group showed the lowest score( P<0.05) while there was no statistical difference between the control group and emotion-medicine group (P>0.05).In saccharose water consumption tests, the consumption of sugar water in PTSD group was the least compared with the other two groups (P<0.05), while there was no statistical difference between the control group and emotion-medicine group (P>00.5) .Conclusion The negative influence derived from maternal PTSD could be handed down to the offsprings resulting in growth and development retardation.Kidney-supplementing remedy and emotional nursing could recover the retardation and even promote growth and development.Our findings might lay a founda-tion for further study on the genetic mechanism and treatment and prevention facing PTSD fetus injury.
