清肺通络膏及其拆方对大鼠呼吸道合胞病毒肺炎外周血T细胞亚群及相关细胞因子的影响

         

摘要

目的 观察清肺通络膏及其拆方对呼吸道合胞病毒肺炎大鼠血T细胞亚群及相关细胞因子的影响,探讨其作用机制.方法 将30只Wistar大鼠随机分为正常组、模型组、全方组、主药组、辅药组,每组6只.除正常组外,其余组以呼吸道合胞病毒滴鼻感染大鼠,然后备给药组分别予清肺通络膏全方、主药和辅药敷背.干预后第7天,应用流式细胞仪测定外周血T细胞亚群,应用ELISA法检测外周血白细胞介素-4(IL-4)、干扰素-γ(INF-γ)水平.结果 模型组CD3+、CD3+ CD8+及INF-γ水平均明显低于正常组(P均<0.05),CD4 +/CD8+及IL-4水平均明显高于正常组(P均<0.05);各给药组CD3+、CD3+ CD8+及INF-γ水平均明显高于模型组(P均<0.05),CD4 +/CD8+及IL-4水平均明显低于模型组(P均<0.05),且全方组上述各指标改善情况均明显优于主药组、辅药组(P均<0.05).结论 呼吸道合胞病毒感染大鼠存在T细胞亚群及相关细胞因子的失衡,清肺通络膏能调整其失衡,且全方效果更佳.%Objective It is to observe the influence of Qingfeitongluo-pastes and its recipe composition on T-lymphocyte subsets and related cytokines in peripheral blood of rats with respiratory syncytial virus pneumonia,and to explore its mechanism.Methods 30 Wistar rats were randomly divided into normal group,model group,whole decoction group,main herb group and adjuvant herb group,each group had 6 rats.The rats in all the groups except normal group were infected with respiratory syncytial virus by nasal drip and were treated with respective medicine by external application on their back.After intervention for 7 days,the levels of T-lymphocyte subset in peripheral blood were measured by flow cytometry and related cytokines in peripheral blood including IL-4,INF-γ were measured by ELISA.Results The levels of CD3 +,CD3 + CD8 + T and INF-γin model group were significantly lower,but CD4+/CD8 + T and IL-4 were higher than those in normal group(P <0.05).The levels of CD3 +,CD3 + CD8 + T and INF-γin every treatment group were significantly higher,but CD4 +/CD8 + T and IL-4 were lower than those in model group(P <0.05),furthermore,the improvements of these indexes in the whole decoction group were better than those in main herb group and adjuvant group (P < 0.05).Conclusion T-lymphocyte subset and related cytokines were out-of-balance when the rats were infected by respiratory syncytial virus.Qingfeitongluo-pastes can adjust its imbalance,and the effect of its whole decoction was the best (P < 0.05).
