温阳振衰颗粒对阿霉素致心肌细胞损伤MEF2表达的影响

     

摘要

目的 观察温阳振衰颗粒含药血清对H9 C2心肌细胞损伤模型肌细胞增强因子2(MEF2)表达的影响.方法 实验分为正常对照组、H9C2损伤组、5%含药血清组和10%含药血清组.H9C2损伤组的细胞培养基中加入2.67μmol/L的阿霉素(ADR),两含药血清组培养基中加入2.67μmol/L的ADR和5%,10%温阳振衰颗粒含药血清,共培养44 h.检测实验末各组H9C2心肌细胞中p-MEF2、MEF2表达量.结果 各损伤组细胞存活率和p-MEF2表达量均明显低于正常对照组(P均<0.05),5%含药血清组和10%含药血清组细胞存活率和p-MEF2表达量均明显高于H9C2损伤组(P均<0.05),且10%含药血清组均明显高于5%含药血清组(P均<0.05);各组间H9C2心肌细胞MEF2表达量比较差异均无统计学意义(P均>0.05).结论 温阳振衰颗粒能抑制MEF2蛋白的过度去磷酸化,这可能是其治疗慢性心力衰竭的重要机制之一.%Objective It is to observe the effect of Wenyang Zhenshuai Granule on expression of myocyte enhancer fac-tor 2 (MEF2) in adriamycin induced H9C2 myocardial cell damage model. Methods H9C2 myocardial cells were divided into normal control group, H9C2 damage group, 5% drug serum group and 10% drug serum group. In the H9C2 damage group, 2. 67 mol/L of adriamycin (ADR) was added to the cell culture medium; in the two drug serum group was added with 2. 67 mol/L ADR and 5%, 10% drug serum of Wenyang Zhenshuai Granule, and co cultured for 44 hours. The ex-pression levels of p-MEF2 and MEF2 in H9C2 myocardial cells were detected at the end of the experiment. Results The cell survival rate and p-MEF2 expression level in each damage group were significantly lower than those in the normal con-trol group (all P<0. 05), the cell survival rate and p-MEF2 expression level in the 5% drug serum group and the 10%drug serum group were significantly higher than those in the H9C2 damage group (all P<0. 05), and those of the 10%drug serum group were significantly higher than the 5% drug serum group (all P<0. 05). There was no significant differ-ence in the expression of MEF2 between H9C2 cells in all groups (P>0. 05). Conclusion Wenyang Zhenshuai Granule can inhibit the excessive dephosphorylation of MEF2 protein, this may be one of the important mechanisms in the treatment of chronic heart failure.
