复方甘草甜素脂质体对人肝星状细胞凋亡和增殖的作用

         

摘要

Objective To observe the effects of compound glycyrrhizin liposome on the apoptosis and proliferation of human hepatic stellate cells ( HSCs),and to explore a new kind of pharmaceutical form on the treatment of hepatic fibrosis. Methods HSCs were incubated with compound glycyrrhizin liposome, or com-pound glycyrrhizin, or liposome alone, or blank for 24 hours. The apoptotic rate of HSCs was tested by Flow cytometry, and the proliferative inhibition rate of the HSCs was determined by MTT assay. Results 1. The apoptotic rates of the HSCs induced with compound glycyrrhizin liposome and compound glycyrrhizin were significantly higher than that of two control groups. The apoptotic rate of the HSCs induced with compound glycyrrhizin liposome was higher than that with the same concentration the compound glycyrrhizin group (P<0.05). 2. The proliferative inhibition rates of the HSCs were induced with compound glycyrrhizin and compound glycyrrhizin liposome were higher than the other two control groups. The proliferative inhibi-tion rate of the compound glycyrrhizin liposome group was increased than the compound glycyrrhizin group in a dose-dependent manner (P<0.05). There was no significant difference between pure HSCs control group and liposome group (P>0.05). Conclusion The compound glycyrrhizin liposome is of stronger effects of inducing apoptosis and inhibition of proliferation in HSCs vitro.%  目的观察复方甘草甜素脂质体对体外培养人肝星状细胞(hepatic stellate cells,HSCs)凋亡和增值的作用,探讨抗肝纤维化新药物剂型。方法体外培养人HSCs,分组:①HSCs对照组;②脂质体对照组;③复方甘草甜素对照组和对应浓度的复方甘草甜素脂质体组。体外培养24 h,用流式细胞术检测HSCs细胞的凋亡率,以及MTT法检测HSCs细胞增殖抑制率。结果①复方甘草甜素脂质体诱导HSC凋亡率高于相同浓度的复方甘草甜素对照组,前两组的HSC凋亡率明显高于脂质体对照组和HSCs对照组。②复方甘草甜素及其脂质体作用后的HSCs增殖的抑制率明显高于两个对照组,复方甘草甜素脂质体组高于复方甘草甜素对照组(P<0.05),抑制率的递增呈浓度依赖性;HSCs对照组和脂质体对照组无明显差别(P>0.05)。结论复方甘草甜素脂质体促进HSCs凋亡及抑制HSCs增殖的作用较复方甘草甜素明显增强,单纯脂质体对体外培养的HSCs的凋亡和增殖无明显影响。
