非结核分枝杆菌临床株rpoB基因多态性分析

摘要

目的：分析常见非结核分枝杆菌(non-tuberculous mycobacteria，NTM)临床株 rpoB 基因的多态性。方法提取NTM 标准菌株和临床菌株的 DNA，扩增其 rpoB 基因部分序列并进行多态性分析。结果10种共27株 NTM 临床株中有8种共14株的 rpoB 基因存在多态性位点，其中脓肿分枝杆菌和耻垢分枝杆菌中均发现3个多态性位点。结论NTM 临床菌株的 rpoB 基因存在多态性，是否会影响其在 NTM 分子鉴定中的应用仍需进一步研究。%Objective To analysis the polymorphism of ropB gene in clinical isolates of common non-tuberculous mycobacteria (NTM). Methods DNA extraction was performed on reference strains and clinical isolates of NTM. Then amplification, sequencing and polymorphism analysis of rpoB gene were accomplished. Results In 27 clinical isolates from 10 NTM species, 14 isolates from 8 species harbored polymorphic condon of ropB gene. Three polymorphic dondons were found in ropB gene of clinical isolates of both Mycobacterium abscessus and Mycobacterium smegmatis.Conclusion Polymorphism of ropB gene was found in clinical isolates of NTM and further study is needed to demonstrate the influence of ropB gene polymorphism on molecular identification of NTM.
