腰痛宁胶囊有效部位组合对 IL-1β诱导大鼠滑膜细胞增殖及 IL-6的影响

摘要

目的：研究腰痛宁胶囊组方有效部位的不同组合对 IL-1β诱导的大鼠滑膜细胞异常增殖和对细胞因子 IL-6分泌的影响，以及各有效部位之间的相互作用关系。方法：大鼠滑膜细胞株 RSC-364体外培养传至第2代后，加入腰痛宁拆方及全方共计6个腰痛宁有效部位组合样品，培养48 h，采用 CCK-8以及 ELISA 法分别测定滑膜细胞增殖及 IL-6分泌水平。结果：6个腰痛宁拆方及组方样品均能够抑制滑膜细胞异常增殖以及 IL-6的分泌，腰痛宁全方的综合活性最佳且其有效部位间存在很强的协同作用。结论：腰痛宁胶囊可有效抑制大鼠滑膜炎，对滑膜细胞具有一定保护作用，腰痛宁胶囊的药引黄酒对于腰痛宁处方中有效部位的细胞活性有重要的促进作用。但可在保证药效前提下减少组方中药或有效部位种类对腰痛宁处方进行精简优化，以增强其质量可控性与安全性。%Objective:To evaluate the in vitro effects and interactions of active fractions in Yaotongning Capsule (YTNC)by detecting proliferation of interleukin-6 (IL-6 )and secretion of rat synovial cell-364 (RSC-364)induced by interleukin-1β(IL-1β).Methods:Six samples including YTNC disassembled prescriptions and the YTNC whole recipe were added to the second generation of culturing RSC-364.After culturing the system for 48 hours,the proliferations of synovial cells and interleukin-6 were examined by cell counting kit-8 (CCK8)assay and enzyme linked immunosorbent assay (ELISA), respectively.Results:The YTNC whole recipe and YTNC disassembled prescriptions successfully inhibited abnormal proliferation of IL-1β-induced synovial cells,as well as the secretion of IL-6.The YTNC whole recipe showed the best integrated activity among the six samples,and strong synergistic effect was observed among the active fractions of the YTNC whole recipe.Conclusion:YTNC prescription can inhibit rat synovitis deterioration and protect synovial cells to some degree.Moreover,the effects of the combination of YTNC material medicines’active fractions on the investigated cell activities are obviously stimulated by YTNC’s vehicle,Chinese rice wine.However,in order to improve quality controllability and safety of YTNC without decreasing its efficacy,the categories of material medicines and active fractions in YTNC could still be reduced to get more simple prescriptions.
