白细胞介素6信号转导通路与糖尿病肾病的关系

         

摘要

Diabetic nephropathy(DN) is a leading cause of chronic kidney disease .Interleukin-6(IL-6) signaling par-ticipates in inflammation responses central to the progression of DN .Current evidence suggests that these IL-6 responses are mediated via gp130-STAT-3 dependent mechanisms which ,on one hand ,trigger globally the transition from innate to adap-tive immune response ,and on the other hand act locally for tissue remodeling and immune cell infiltration .In DN conditions the role of IL-6 is not well elucidated .IL-6 classical signaling pathway via receptor mbIL-6R and IL-6 cross-signaling path-way via soluble IL-6R are proven to participate in the pathogenesis and progression of DN .%糖尿病肾病(DN)是慢性肾脏病的主要原因.白细胞介素6(IL-6)信号转导通路参与DN进展的炎症反应.目前的研究表明,这些IL-6应答是通过依赖糖蛋白130-信号转导与转录活化因子3机制介导的,该机制一方面触发从先天性到适应性免疫应答的转变,另一方面用于局部组织重塑和免疫细胞浸润.而在DN中,IL-6的作用并没有得到很好的阐明.IL-6介导的经典信号转导通路通过IL-6结合受体蛋白和IL-6介导的跨信号转导通路通过可溶性IL-6受体被证明参与DN的发病机制.
