多囊卵巢综合征模型大鼠血清生长素水平及卵巢组织生长素的表达及意义

         

摘要

Objectives The present study investigated the relationship between ghrelin and the estrous cycle. The expression and significance of ghrelin in the ovary and endometrium of rats models of polycystic ovary syndrome (PCOS) were also studied. Methods A total of 44 eight-week-old SD rats were selected for the present study. Insulin and human chorionic gonadotrophin (HCG) were used in 24 rats to reproduce PCOS model, whereas the remaining 20 were given similar amount of normal saline as the control group. The immunohistochemical method was used to detect the ghrelin expression in the ovary and endometrium of these two groups, whereas ELISA was adopted to determine serum ghrelin levels during different estrous cycles, and also before and after fasting. Results Significant differences in the serum ghrelin levels were observed before and after fasting in the PCOS rat models (P=0.004). The serum ghrelin levels during different estrous cycles showed periodic variations in the control rats, but without significant difference (P=0.084). The serum ghrelin level of the PCOS model group was lower than that in the control group, but without significant statistical difference (P=0.067). Ghrelin expression could be seen in the ovarian tissue during the whole estrous cycle of rats in the control group. In particular, expression was significantly elevated at metestrus. The ghrelin expression in the model group was significantly reduced compared with the control group during different estrous cycles (P=0.03l). No ghrelin expression was found in the endometrium of rats in both groups. Conclusions Ghrelin expression may fluctuate periodically during the estrous cycle, and its highest level occurs at the metestrus. Ghrelin may be related to the corpus luteum function, and has no evident relation with the endometrial function.%目的 研究大鼠血清生长素(ghrelin)与动情周期的关系,以及ghrelin在多囊卵巢综合征(PCOS)模型大鼠的卵巢及子宫内膜中的表达及意义.方法 选择8周龄SD雌性大鼠44只,其中24只利用胰岛素联合人绒毛膜促性腺激素造模,作为PCOS造模组,其余20只给予同等剂量的生理盐水作为对照组.利用免疫组化方法检测两组大鼠卵巢及子宫内膜上ghrelin的表达,酶联免疫法(ELISA)测定不同动情周期及禁食前后大鼠的血清ghrelin水平.结果 PCOS造模组大鼠在禁食前后血清ghrelin水平差异有统计学意义(P=0.004),对照组大鼠不同动情周期的血清ghrelin水平存在周期变化,但差异无统计学意义(P=0.084).PCOS造模组大鼠的血清ghrelin水平较对照组降低,但差异无统计学意义(P=0.067).对照组大鼠不同动情周期的卵巢组织颗粒细胞上均能见到ghrelin表达,在动情后期表达明显增加.与对照组不同动情周期比较,造模组ghrelin的表达明显减少(P=0.031).两组大鼠子宫内膜上均未见到ghrelin表达.结论 大鼠血清ghrelin水平可能随着动情周期变化而变化,以动情后期水平最高,ghrelin可能与卵巢黄体功能有关,而与子宫内膜功能无明显联系.
