黄芪多糖对糖尿病小鼠血清胰岛素样生长因子-1的影响

         

摘要

Objective:To study the influence of astragalus polysaccharides(APS) on insulin-like growth factor(IGF-1) in diabetic mice.Method:A total of 60 mice were randomly divided into three groups, blank group,APS group and control group,each group had 20 cases.The APS group and the control group were fed with diets high in saturated fat and peritoneal injected small dose of Streptozotocin for DM mice model.After modeling, the APS group was intragastrically administrated to APS for 8 weeks, while physiological saline was used as control. The change of the serum level of IGF-1, ICAM-1, VCAM-1 and MCP-1 were observed by enzyme-linked immunosorbent (ELISA),and these indicators were used to evaluate the function of APS.Result:After treatment, the serum level of ICAM-1, VCAM-1 and MCP-1 were significantly declined, while the serum level of IGF-1 increased significantly, these changes were significant differences compared with the control group (P<0.05). Conclusion: Short-term treatment of APS can improve the level of serum IGF-1 in DM mice, accompanied by the decline of inflammatory indexes such as ICAM-1,VCAM-1.%目的:研究黄芪多糖(APS)对糖尿病(DM)模型小鼠血清胰岛素样生长因子-1(IGF-1)的影响及其治疗糖尿病的作用机制。方法:将60只昆明小鼠随机分为空白组、APS组和对照组,每组20只。其中APS组和对照组采用高脂饮食喂养加小剂量链脲佐菌素(STZ)腹腔注射法诱导成DM小鼠模型。造模完成后,APS组给予APS药剂灌胃治疗,对照组予以等体积的生理盐水灌胃治疗,两组治疗均持续8周。采用酶联免疫吸附法(ELISA)检测其血清IGF-1、ICAM-1、VCAM-1及MCP-1水平的变化,观察APS对DM小鼠模型的作用。结果:治疗后,APS组DM小鼠血清ICAM-1、VCAM-1及MCP-1水平与治疗前比较差异均有统计学意义(P<0.05);治疗后,APS组中DM小鼠血清IGF-1水平明显升高,与治疗前比较差异有统计学意义(P<0.05)。结论:短期APS治疗能提高DM小鼠血清IGF-1水平,并伴随着ICAM-1、VCAM-1等炎性指标的血清水平降低。
