谷胱甘肽对糖基化终末产物环境中人脐静脉内皮细胞前列环素和内皮素-1分泌的影响

     

摘要

Aim:To investigate the influence of reduced glutathione ( GSH ) on prostacyclin ( PGI2 ) and endothelin-1 (ET-1) secretion of human umbilical vein endothelial cells ( HUVEC ) cultured with advanced glycation end product (AGE). Methods:The secretion of PGI2 and ET-1 of HUVEC,which were cultured in M199 medium with AGE(0.8 g/ L),with GSH(6 mmol/L),with AGE(0. 8 g/L)and GSH(6 mmoI/L) ,and M199 alone for 24,48 , and 72 h, respectively, was assayed by RIA. Results:AGE could decrease the secretion of PGI2 from HUVEC( at 24,48 .and 72 h,FKGf =61.581, 142.071 and 98.203,P< 0.001), increase the secretion of ET-1 (at 24,48,and 72 h,FAGE =22.927,63.229 and 26. 962, P< 0.001 ). The decrease and increase could be diminished by GSH ( PGI2: at 48 and 72 h, Finteraction=8. 129, 6. 857,and 6. 857,P<0.05;ET-1:at 48 and 72 h,Finteraction = 105. 892 and 48. 258,P <0. 001 ). Conclusion: GSH could protect vascular endothelial cells from the injury induced by AGE.%目的:了解谷胱甘肽(GSH)对糖基化终末产物(AGE)环境中人脐静脉内皮细胞(HUVEC)前列环素(PGI2)和内皮素-1(ET-1)分泌的影响。方法:分别用含M199培养基、含AGE(0.8g/L)的M199培养基、含GSH(6mmol/L)的M199培养基及含AGE(0.8g/L)和GSH(6 mmol/L)的M199培养基培养HUVEC,培养24、48、72 h后测定各组细胞中PGI2和ET-1的分泌量。结果:AGE可显著降低HUVEC的PGI2分泌量(24、48、72 h时的FAGE=61.58l,142.07l和98.203,P均<0.001),升高ET-1分泌量(24、48、72 h时的FAGE=22.927,63.229和26.962,P均<0.001);GSH可拮抗上述作用[(PGl2:48、72 h时的F交互=8.12l和6.857,P均<0.05;ET-l:48、72 h时的F交互=105.892和49.257,P均<0.001)]。结论:GSH可减轻AGE对血管内皮细胞的损伤。
