黄连素对肿瘤坏死因子TNF-α介导的炎症作用机理研究

         

摘要

[Objective] To investigate whether berberine could block tumor necrosis factor-α ( TNF-α) induced inflammation in HU-VEC. [Methods] HUVEC were treated with TNF-α(1μg/L) and /or berberine (0. Lmg/L, 0. 5mg/L, lmg/L, 2mg/L) for 16 hours. The mRNA level of vascular cell adhesion molecule-1 (VCAM-1) was determined by real-time PCR. Human macrophage THP-1 adhereing to HUVEC was measured by rose Bengal staining. Migration function and MTT text were used to demonstrate the effects of berberine on the smooth muscle cell's migration and proliferation functions . [Results] TNF-α treatment significantly increased the mRNA expression of VCAM-1 in HUVEC,as compared to controls berberine pretreatment blocked the TNF-a induce mRNA expression of VCAM-1 ,and also effectively blocked THP-1 adhesion. Further more,berberine inhibited TNF-a induced contractile fiber cell's migration and proliferation functions. [Conclusion] These data suggested that berberine might have potential therapeutic effects in controlling inflammation in vascular diseases.%[目的]探讨黄连素抑制HUVEC中TNF-α引起的细胞炎症的发生。[方法]分别培养HUVEC和A7r5细胞,不同药物浓度处理后,提取HUVEC mRNA,用实时荧光定量PCR测定VCAM-1表达水平;用虎红染色法检测HUVEC和人单核细胞(THP-1)的黏附性;用细胞迁移和MTT增殖实验分别检测黄连素对A7r5迁移和增殖作用的影响。[结果]黄连素预处理能有效阻止TNF-α诱导的VCAM-1mRNA增高,并减弱由其导致的THP-1对HUVEC的黏附性增加,同时抑制由TNF-α诱导的平滑肌细胞的迁移和增值作用。[结论]黄连素对炎症及心血管疾病有潜在的治疗作用。
