复方菊明提取物对高血压伴高尿酸血症模型大鼠尿酸的影响

         

摘要

[Objective] To observe the effect and mechanism of CJE on uric acid level in spontaneously hypertensive rats accompanied with hyperuricemia, which induced by high-purine diet. [Methods] Spontaneously hypertensive rats were continuously received high-purine diet for 8 weeks to establish hyperuricemia and hypertension model. CJE (10.0, 5.0, and 2.5g·kg-1) were given once a day for 8 weeks. At0, 3, 6 and 8 weeks, blood sample was collected to detect levels of serum uric acid (UA), nitrogen (BUN), and creatinine (Cr). After the last administration, serum was collected to detect xanthine oxidase (XOD) and adenosine deaminase(ADA) activity. [Results] All CJE(10.0g/kg, 5.0g/kg, 2.5g/kg)can reduce serum UA, BUN, Cr levels and serum XOD, ADA activities. [Conclusion] CJE could significantly decrease uric acid level and improve kidney function. The mechanism may be related with the inhibition of XOD and ADA activity.%[目的]观察复方菊明提取物(Compound Juming extract,CJE)对高嘌呤饲料饲喂自发性高血压大鼠(Spontaneously Hypertensive Rats,SHR)致高血压伴高尿酸血症模型大鼠的降尿酸作用,并初步探讨其作用机制.[方法]采用SHR连续饲喂高嘌呤饲料8周建立高血压伴高尿酸血症模型大鼠,在造模的同时灌胃给予CJE,连续8周.分别于给药前第0周,给药第3周、第6周、第8周取血测定血清尿酸(uric acid,UA)、肌酐(creatinine,Cr)、尿素氮(nitrogen,BUN)水平;末次给药后取血测定血清黄嘌呤氧化酶(xanthine oxidase,XOD)、腺苷脱氨酶(adenosine deaminase,ADA)活性.[结果]CJE(10.0、5.0、2.5g·kg-1)均能降低高血压伴高尿酸血症大鼠血清UA、Cr、BUN水平,降低血清XOD、ADA活性.[结论]CJE具有显著的降尿酸及改善肾功能作用,其机制可能与抑制XOD和ADA活性有关.
