金银花水煎剂对D-半乳糖致衰老模型小鼠的抗氧化作用

         

摘要

[目的]探讨金银花水煎剂对D-半乳糖致衰模型小鼠的抗氧化作用。[方法](1)取一月龄ICR小鼠50只,随机分为对照组、衰老组、金银花高、中、低剂量组。D-半乳糖构建衰老模型过程中,金银花高(10g·kg-1)、中(5g·kg-1)、低(2.5g·kg-1)剂量组每天按剂量分别灌胃。对照组和衰老组灌等量的生理盐水。连续6W后采集小鼠血清样品和各组织匀浆样品进行生化指标SOD、MDA、GSH-Px检测。[结果]42d后,衰老组小鼠出现不同程度的行动缓慢,皮毛光泽暗淡;体重低于正常组;血清、肝、肾、大脑组织中的SOD值、GSH-Px水平低于正常组;血清、肝、肾组织中的MDA含量均高于正常组(P<0.05或P<0.01);以上数据说明造模基本成功。与衰老组比较,金银花给药组各项指标明显改善,金银花给药组体重增加高于衰老组(P<0.01);血清、肝、肾SOD值及GSH-Px水平均高于衰老组(P<0.05或P<0.01),MDA含量低于衰老组(P<0.05或P<0.01)。[结论]金银花对D-半乳糖致衰老小鼠具有抗氧化作用,且中剂量组(5g·kg-1)效果显著。%[Objective]To study the antioxidant effect of Lonicera japonica extract in D-galactose induced aging model mice. [Methods] Fifty 1-month-old ICR mice were randomly divided into 5 groups: control group, aging group, Lonicera japonica high, medium and low dose groups. During building the aging model, the Lonicera japonica high(10g·kg-1),medium(5g·kg-1) and low(2.5g ·kg-1 ) dosage by gavage in Lonicera japonica treatment groups with Lonicera japonica extract every day. Control group and aging group treated with the same amount of normal saline. 6 weeks later, col ected mice serum samples and various tissue samples for biochemical indicators of SOD, MDA, GSH-Px detection. [Results]After 42days, it appeared different degrees of moving slowly and fur tarnish in aging mice. Aging mice group weight was less than the normal group. Serum, brain, liver and kidney tissue SOD activity, GSH-Px levels were lower than the normal group; serum, liver and kidney tissue MDA levels were higher than the normal group( P <0.05 or P<0.01);These data il ustrated the aging model was general y successful. Compared with the aging mice group, the indicators of Lonicera japonica treatment groups had been improved significantly. Lonicera japonica treatment groups weight were higher than the aging group( P<0.01);serum, liver, kidney SOD activities and GSH-Px levels were higher than the aging group(P<0.05 or P<0.01);MDA levels were lower than the aging group(P<0.05 or P<0.01).[Conclusion] Lonicera japonica extract could resist the oxidation process in D-galactose induced aging model mice, and the middle dose group(5g·kg-1) effect was remarkable.
