从氧化应激角度探讨清肾颗粒对肾间质纤维化大鼠的干预作用

         

摘要

[Objective] Through the detection of unilateral ureteral obstruction(UUO) induced renal interstitial fibrosis(RIF) in the renal tissue of rats reactive oxygen species(ROS), malondialdehyde(MDA) and superoxide dismutase(SOD) expression level to explore the intervention of Qingshen granule with oxidative stress. [Method] Used unilateral ureteral obstruction method(UUO) to model rats, 60 SD rats randomly divided into Qingshen granule group(n=12), candecartan cilexetil group(n=12), model group(n=13), sham operation group(n=12) and normal group(n=11). Due to infection, injury, surgical anesthetic overdose deaths in 12 rats, and finally into the data analysis of 48 rats, Qingshen granule group(n=10), candecartan cilexetil group(n=10), model group(n=8), sham operation group(n=9) and normal group(n=11) Qingshen granule group were given Qingshen granule 0.4g(100g·d) gavage, candecartan cilexetil group were given candecartan cilexetil capsule 0.05g/(100g·d) gavage, model group, sham operation group and the normal group were given the same amount of normal saline per day orally, for 4 weeks. The rats serum creatinine(Scr), blood urea nitrogen(BUN) and 24h urinary protein(U-Pro/24h) level were detected. Using enzyme-linked immunosorbent assay(ELISA) to detect the renal tissue of rats ROS, MDA and SOD. [Results] Qingshen granule group, candecartan cilexetil group, model group BUN, Scr, U-Pro/24h, ROS and MDA were higher compared with normal group and sham operation group( P<0.01), the level of SOD was lower(P<0.01). Compared with model group, Qingshen granule group and candecartan cilexetil group, the levels BUN, Scr, U-Pro/24h, ROS and MDA were significantly lower(P<0.01), the level of SOD was higher(P<0.01), Qingshen granule group was better than the candecartan cilexetil group(P<0.05). [Conclusion] Qingshen granule can reduce the renal tissue of ROS and MDA, rise the renal tissue of SOD, relieve oxidative stress of kidney and prevent or mitigate the occurrence and development of renal fibrosis.%[目的]观察肾间质纤维化(renal interstitial fibrosis,RIF)大鼠肾组织中活性氧(reactive oxygen species,ROS)、丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)的表达,探讨清肾颗粒对RIF大鼠氧化应激的影响.[方法]采用单侧输尿管梗阻(unilateral ureteral obstruction,UUO)建立RIF模型,将60只SD大鼠随机分为清肾颗粒组(n=12)、坎地沙坦组(n=12)、模型组(n=13)、假手术组(n=12)和正常组(n=11).因感染、麻醉药过量、手术损伤等原因死亡12只大鼠,最后进入数据分析有48只大鼠,即清肾颗粒组(n=10)、坎地沙坦组(n=10)、模型组(n=8)、假手术组(n=9)和正常组(n=11).清肾颗粒组予清肾颗粒水溶液0.4g/(100g·d)灌胃,坎地沙坦组予坎地沙坦水溶液0.05g/(100g·d)灌胃,模型组、假手术组及正常组每日给予等量生理盐水灌胃,疗程均为4周.检测各组大鼠血尿素氮(blood urea nitrogen,BUN)、肌酐(serum creatinine,Scr)和24h尿蛋白定量(24h urinary protein,U-Pro/24h),采用双抗体夹心酶联免疫吸附(enzyme linked immunosorbent assay,ELISA)法检测大鼠肾组织中ROS、MDA、SOD含量.[结果]清肾颗粒组、坎地沙坦组、模型组BUN、Scr、U-Pro/24h、ROS、MDA水平均高于正常组和假手术组(P<0.01),SOD水平均低于正常组和假手术组(P<0.01);与模型组比较,清肾颗粒组和坎地沙坦组BUN、Scr、U-Pro/24h、ROS、MDA水平明显降低(P<0.01),SOD水平明显升高(P<0.01),清肾颗粒组优于坎地沙坦组(P<0.05).[结论]清肾颗粒可以降低UUO大鼠肾组织中ROS、MDA,升高SOD水平,减轻肾组织中氧化应激反应,提高抗氧化能力,进而发挥抗RIF作用.
