姜黄素调控 Bcl -2和 Bax 蛋白表达减轻胃缺血/再灌注损伤

     

摘要

Objective To investigate the protective effects of curcumin pretreatment on the gastric ischemia/reperfu-sion (GI/R) injury in rats.Methods A rat model of GI/R injury was established through occlusion of the celiac artery in rats for 30 min followed by reperfusion for 60 min.Then the area of damaged gastric mucosa was calculated by soft-ware, while the index of gastric mucosal damages was assessed according to the white grading method.The amounts of Bcl -2 and Bax in the tissues of gastric mucosa were detected using immunohistochemical methods and image analysis. Results Compared with the control, rats in the GI/R group presented damages over remarkably larger areas and higher damage indices (P <0.05 or P <0.01).Curcumin pretreatment could relieve the gastric mucosa injury caused by GI/R. The area of gastric mucosal damages and the damage index became significantly lowered with the rising of the concentra-tions of curcumin (P <0.05 or P <0.01).Meanwhile, curcumin pretreatment could weaken the amounts of Bax but en-hance the level of Bcl -2 in the gastric mucosal tissues after GI/R in a dose -dependent manner (P <0.05 or P <0.01).Conclusion Curcumin pretreatment can protect the gastric mucosa and attenuate its damages caused by GI/R through enhancing Bcl -2 but weakening Bax protein expression.This finding indicates the potential role of curcumin as a protective agent against organ injuries caused by ischemia/reperfusion.%目的:观察姜黄素预处理对大鼠胃缺血/再灌注(gastric ischemia -reperfusion,GI/R)损伤的保护作用。方法采用夹闭大鼠腹腔动脉30 min 再灌注1 h 建立 GI/R 损伤模型。应用计算机软件分析计算 GI/R 引起的胃黏膜损伤面积,根据 whtie 评分法计算胃黏膜损伤指数,用免疫组化法与图像分析技术检测胃黏膜组织 B 细胞淋巴瘤因子2(Bcl -2)与 Bcl -2相关 X 蛋白(Bax)表达量。结果与对照组相比,GI/R 组胃黏膜损伤面积和损伤指数明显增加(P <0.05,P <0.01);姜黄素预处理能够有效减轻 GI/R 引起的胃黏膜损伤,随着姜黄素浓度的增加,GI/R 引起的胃黏膜损伤面积和损伤指数显著减少,并具有浓度依赖性(P <0.05,P <0.01)。同时,姜黄素预处理能够降低 GI/R 引起的胃黏膜组织 Bax 蛋白表达,增加胃黏膜组织 Bcl -2蛋白表达,且随着姜黄素浓度的增加,其差异具有浓度依赖性(P <0.05,P <0.01)。结论姜黄素预处理通过增加抗凋亡蛋白 Bcl -2、减少促凋亡蛋白 Bax 的表达有效抑制 GI/R 引起的胃黏膜损伤,对胃黏膜起到一定程度的保护作用,对临床缺血/再灌注引起的器官损伤具有潜在的治疗作用。
