长期培养外周血单个核细胞的简易分离纯化方法

摘要

目的 对长期培养的外周血单个核细胞(PBMC)进行纯化,为后续研究提供方便.方法 用聚蔗糖-泛影葡胺密度梯度离心法对长期培养的PBMC进行再次分离和纯化.结果 用聚蔗糖-泛影葡胺密度梯度离心法再次处理长期培养的PBMC,可以得到纯度较高的PBMC.结论 聚蔗糖-泛影葡胺密度梯度离心法是纯化长期培养的PB-MC的一种简便、快捷、成本低的方法之一.%Objective To purify the long-term cultured peripheral blood mononuclear cells(PBMC) and provide convenience for the subsequent research. Methods The long-term cultured PBMC were isolated and purified by Ficoll-Hypaque density gradient centrifugation. Results After the above centrifugation, a higher purity of PBMC can be easily obtained. Conclusion The Ficoll-Hypaque density gradient centrifugation is a simple,fast,low cost method for purifying long-term cultured PBMC.
