磷脂酸磷酸酶2域1A在不同转移能力肝癌细胞系中的表达

Objective To investigate the difference in the expression of phosphatidic acid phosphatase type 2 domain containing 1A (PPAPDC1 A) in hepatocellular carcinoma cell lines with different metastatic ability.Methods The expression of PPAPDC1A protein and mRNA in seven hepatocellular carcinoma cell lines(BEL-7402,HepG2,MHCC-97L,SMMC-7721,HCCL-M6,BEL-7404,MHCC-97H cell) with different metastatic ability was detected by Western blot and real-time fluorescent quantitative polymerase chain reaction,respectively.Results The expression order of PPAPDC1A mRNA and PPAP-DC1A protein in the seven different metastatic hepatocellular carcinoma cells was BEL-7402 cell ＜ HepG2 cell ＜ MHCC-97L cell ＜ SMMC-7721 cell ＜ HCCL-M6 cell ＜ BEL-7404 Cell ＜ MHCC-97H cell.There was significant difference in the expression of PPAPDC1A mRNA and PPAPDC1A protein in the seven liver cancer cells with different metastatic ability (F =38.373,441.937;P ＜ 0.05).The expression of PPAPDC1A protein in HepG2 cell was significantly higher than that in BEL-7402 cell (P ＜ 0.05).The expression of PPAPDC1A protein in MHCC-97L cell was significantly higher than that in HepG2 cell (P ＜0.05).The expression of PPAPDC1A protein in SMCC-7721 cell and HCCL-M6 cell was significantly higher than that in MHCC-97L cell (P ＜ 0.05).The expression of PPAPDC1A protein in HCCL-M6 cell was significantly higher than that in SMCC-7721 cell (P ＜0.05).The expression of PPAPDC1A mRNA and PPAPDC1A protein in MHCC-97L,SMCC-7721,HCCL-M6,BEL-7404 and MHCC-97H cell was significantly higher than that in BEL-7402 cell (P ＜ 0.05).The expression of PPAPDC1A mRNA and PPAPDC1A protein in SMCC-7721,HCCL-M6,BEL-74041 and MHCC-97H cell was significantly higher than that in HepG2 cell (P ＜0.05).The expression of PPAPDC1A mRNA and PPAPDC1A protein in BEL-7404 and MHCC-97H cell was significantly higher than that in MHCC-97L cell (P ＜ 0.05).The expression of PPAPDC1A mRNA and PPAPDC1A protein in BEL-7404 and MHCC-97H cell was significantly higher than that in SMCC-7721 cell (P ＜ 0.05).The expression of PPAPDC1A mRNA and PPAPDC1A protein in BEL-7404 and MHCC-97H cell was significantly higher than that in SMCC-7721 cell (P ＜ 0.05).The expression of PPAPDC1A mRNA and PPAPDC1A protein in BEL-7404 and MHCC-97H cell was significantly higher than that in HCCL-M6 cell (P ＜ 0.05).The expression of PPAPDC1A mRNA and PPAPDC1A protein in MHCC-97H cell was significantly higher than that in BEL-7404 cell (P ＜ 0.05).Conclusion PPAPDC1A may promote the invasion and metastasis of hepatocellular carcinoma.%目的 研究磷脂酸磷酸酶2域1A(PPAPDC1A)在7种不同转移能力的肝癌细胞系中表达的差异.方法 以7种不同转移能力的肝癌细胞系(BEL-7402、HepG2、MHCC-97L、SMMC-7721、HCCL-M6、BEL-7404、MHCC-97H细胞)为研究对象,通过实时荧光定量聚合酶链式反应和Western blot法分别检测其PPAPDC1A mRNA和蛋白的表达.结果 在7种不同转移能力的肝癌细胞中,PPAPDC1A mRNA与PPAPDC1A蛋白的表达从低到高依次为BEL-7402细胞＜ HepG2细胞＜MHCC-97L细胞＜SMMC-7721细胞＜HCCL-M6细胞＜BEL-7404细胞＜MHCC-97H细胞;7种不同转移能力肝癌细胞中PPAPDC1 A mRNA和PPAPDC1A蛋白的表达比较差异均有统计学意义(F=38.373、441.937,P＜0.05).其中HepG2细胞中PPAPDC1 A蛋白的表达显著高于BEL-7402细胞(P＜0.05);MHCC-97L细胞中PPAP-DC1A蛋白的表达显著高于HepG2细胞(P＜0.05);SMCC-7721、HCCL-M6细胞中PPAPDC1A蛋白的表达显著高于MHCC-97L细胞(P＜0.05);IICCL-M6细胞中PPAPDC1A蛋白的表达显著高于SMCC-7721细胞(p＜0.05);MHCC-97L、SMCC-7721、HCCL-M6、BEL-7404、MHCC-97H细胞中PPAPDC1A mRNA和PPAPDC1A蛋白的表达显著高于BEL-7402细胞(P＜0.05);SMCC-7721、HCCL-M6、BEL-7404、MHCC-97H细胞中PPAPDC1A mRNA和PPAPDC1A蛋白的表达显著高于HepG2细胞(P＜0.05);BEL-7404、MHCC-97H细胞中PPAPDC1A mRNA和PPAPDC1 A蛋白的表达显著高于MHCC-97L细胞(P＜0.05);BEL-7404、MHCC-97H细胞中PPAPDC1A mRNA和PPAPDC1A蛋白的表达显著高于SMCC-7721细胞(P＜0.05);BEL-7404、MHCC-97H细胞中PPAPDC1A mRNA和PPAPDC1A蛋白的表达显著高于HCCL-M6细胞(P＜0.05);MHCC-97H细胞中PPAPDC1A mRNA和PPAPDC1A蛋白的表达显著高于BEL-7404细胞(P＜0.05).结论 PPAPDC1A可能促进肝细胞癌的侵袭与转移.
