藏茴香水提物对高脂血症大鼠血脂及肝功能的影响

         

摘要

目的:研究藏茴香水提物对大鼠血脂调节及肝脏的保护作用。方法将10 w 龄雄性 SD 大鼠48只,除空白对照组8只大鼠以外其余各组大鼠均采用高脂饲料喂养8 w 后,断尾采血测血脂,确定造模成功后随机分为模型对照组、血脂康对照组、空白对照组及藏茴香水提物高、中、低剂量组(300、150、75 mg·kg-1·d-1),连续灌胃给药6 w,1次/d,模型对照组与空白对照组灌胃给予等量的生理盐水。每2周测1次体质量;末次给药后腹主动脉采血,测血清中的高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、总胆固醇(TC)、甘油三酯(TG)等血脂水平,测定谷草转氨酶(AST)、谷丙转氨酶(ALT)等脂类酶的活性及谷胱甘肽过氧化酶(GSH-Px)、超氧化物歧化酶(SOD)、脂肪过氧化物丙二醛(MDA)等抗氧化能力的变化。结果与空白对照组比较,模型对照组 TG、TC、LDL-C 均有显著升高,HDL-C 显著降低,差异均有统计学意义(P <0.05)。藏茴香各剂量组 TC、TG、LDL-C 逐渐降低,HDL-C 逐渐升高(说明藏茴香干预能有效调节血脂水平)。与模型对照组比较,藏茴香各剂量组大鼠血清血脂水平差异均有统计学意义。与空白对照组比较,模型对照组血清 AST、ALT 明显升高,有统计学差异(P <0.05),血脂康对照组及藏茴香水提物各剂量组血清 AST、ALT 均显著降低;与模型对照组比较,藏茴香水提物高、中剂量组有统计学差异(P <0.05~0.01),与小剂量组比较无统计学差异(P >0.05)。与空白对照组比较,模型对照组 SOD、MDA 水平升高,差异有统计学意义(P <0.05);与模型对照组比较,藏茴香水提物高剂量组SOD、MDA 水平和血脂康对照组 SOD 水平差异有统计学意义(P <0.05),与藏茴香水取物中、低剂量组比较无统计学差异(P >0.05)。模型对照组与空白对照组比较 GSH-Px 有统计学差异(P <0.001);随着实验的进行,藏茴香水提物各剂量组 GSH-Px 均明显升高,模型对照组与血脂康对照组及藏茴香水取物高剂量组比较有统计学差异(P <0.05),与中剂量及低剂量组比较无统计学差异(P >0.05)。结论高、中剂量藏茴香水提物可明显调节机体脂质代谢,对预防和治疗高脂血症有一定的应用价值。%Objective To investigate the effect of regulation of the blood lipid and protection of liver func-tion ofCarum carvi L.in a hyperlipidemia rat model and explore its mechanism.Methods After two-week acclimation,48 10-week old male rats were divided into 6 groups randomly,including the model group, Xuezhikang group,high,middle and low doses of CCL groups (300,150,75 mg·kg-1 ·d-1 )and the normal group,8 rats in each group.All the groups except the control group were fed by a high-fat diet for 12 days and then tested the blood lipids from the tail vein.Drugs were given to the hyperlipemia rats by in-tragastric administration for 8 weeks,once daily.The model group and the normal group were given nor-mal saline instead.The body weight was measured every two weeks.The blood was collected from the ab-dominal aorta 12 hours after the last administration,high density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol (LDL-C),total cholesterol (TC),the triglyceride (TG),blood lipid levels and aspartate amino transferase (AST),alanine transaminase (ALT)were determined in serum,lipid per-oxidase (GSH-PX)activity,superoxide dismutase (SOD)and lipid peroxide Malondialdehyde (MDA)lev-els were measured.Results Compared with the control group,TG,TC,LDL-C were significantly in-creased,and HDL-C was significantly decreased in model control group,the difference was statistically significant (P <0.05).TC,TG,LDL-C decreased,HDL-C gradually increased in each Carum carvi L.dose group (Intervention can effectively regulate lipid levels).Compared with the model group,serum lipid lev-els in each dose group were statistically significant (P <0.05).Compared with the control group,serum AST,ALT increased significantly in model control group,with significant differences (P <0.05),AST, ALT decreased significantly in Xuezhikang control group and water extract of each dose group;compared with the model group,AST,ALT were significantly different between high and middle dose groups (P <0.05-0.01),and low-dose group showed no significant difference (P >0.05).Compared with the control group,SOD,MDA levels increased in model control group,with significant differences (P <0.05);SOD, MDA levels of high-dose group and SOD level of Xuezhikan were significant compared with the model group (P <0.05).The level of GSH-Px in model control group and control group were significantly differ-ent (P < 0.05);The level of GSH-Px in model control group and Xuezhikang control group high-dose groups were significantly different (P <0.05),with the middle-dose and low-dose group showed no signif-icant difference (P >0.05).Conclusion High and middle dose of Carum carvi L.water extract can signifi-cantly regulate lipid metabolism,have a certain value on preventation and treatment of hyperlipidemia.
