不同动物皮肤对黄体酮乳膏体外经皮透过性的比较

摘要

Objective To investigate the transdermal permeability of progesterone cream on different ani-mals′skin,and suggest the suitable animal′s skin for prescription screening.Methods Franz diffusion cell and HPLC were used to detect the concentration of progesterone in receiving solution.Results Abdominal skin of rats had the best effect of the transdermal penetration of progesterone;the 24 h-accumulated permeation of pro-gesterone reached the maximum and percutaneous rate was the highest.Conclusion Abdominal skin of rats is the most suitable model for proscription screening of transdermal progesterone delivery system.%目的：考察黄体酮乳膏在不同动物离体皮肤中的透皮吸收性能。方法选用小鼠、大鼠、豚鼠、家兔、比格犬和小型猪的离体皮肤，采用 Franz 扩散池法，用高效液相色谱法测定接收液中的黄体酮含量，并进行不同模型的拟合。结果黄体酮乳膏透皮动力学行为符合一级动力学模型。小鼠腹部皮肤对黄体酮的稳态透皮速率（Js）为（8．8543±0．7411）μg·cm－2·h－1，在6种动物皮肤中最高，与其他5种动物皮肤的稳态透皮速率差异均有统计学意义（P 均＜0．05）。结论选择小鼠腹部皮肤作为体外透皮吸收实验模型时，黄体酮乳膏的透皮吸收效果最好。
