新疆假龙胆乙醇提取物不同极性萃取部位的急性毒性实验

摘要

Objective To investigate the acute toxicity of ethanol extract and different polar parts of Gentianella turkestanorum,and to provide the basis for clinical safety medication.Methods The extract of Gentianella turkestanorum was extracted by refluxing with 95％ethanol as the extraction solvent.Volatilization to until no alcohol can be smiled followed by water suspension.Suspension was further extracted using petroleum ether,ethyl acetate and n-butanol,respectively.then,Xinjiang Gentianella ethanol extracts of different polar extracts was obtained.the maximum dosage method was used for acute toxicity test with 100 ICR mice,which,were randomly divided into 5 groups with each having 20 rats: In blank control group,mice were given equal volume of distilled water,the other groups were gastrically gavaged with the maximum concentration of Xinjiang Gentianella ethanol extract,ethyl acetate,n-butanol and water extract(21.20,16,40,40 g/kg)suspension daily according to the 0.4 mL/10 g body weight.The toxicity and the change of weight of mice at 14th day cases were observed.Results Cream rate of ethanol,petroleum ether,ethyl acetate,n-butanol and an aqueous layer extract of Gentianella turkestanorum was 24.4％,8.20％,4.92％,11.47％and 19.67％,respectively.The maximum dose for gastric gavage of ethanol,water,nbutanol,ethyl acetate extracts of Gentianella turkestanorum was 86.88 g crude drug/kg,203.35 g crude drug/kg,348.73 g crude drug/kg,325.20 g crude drug/kg,respectively.Conclusion Ethanol extracts and different polar parts of Gentianella turkestanorum turned out to be non-toxic and safe.%目的 探究新疆假龙胆乙醇提取物不同极性萃取部位的急性毒性,为临床安全用药提供依据.方法 采用95％乙醇为提取溶剂,加热回流提取新疆假龙胆,合并提取液,浓缩.挥至无醇味加水混悬,混悬液依次用石油醚、乙酸乙酯、正丁醇萃取,分别得到新疆假龙胆乙醇提取物不同极性萃取部位;采用最大给药量法进行急性毒性实验,ICR小鼠100只,雌雄各半,按体质量随机均分为5组,每组20只,空白对照组小鼠灌胃给予等容积蒸馏水,其余组分别给予最大浓度、最大体积的新疆假龙胆乙醇提取物、乙酸乙酯、正丁醇、水层萃取部位(21.20、16.00、40.00、40.00 g/kg体质量)混悬液,按0.4 mL/10 g体质量灌胃1次/d,在14 d内观察小鼠的毒性反应及体质量变化情况.结果 新疆假龙胆乙醇提取物、石油醚、乙酸乙酯、正丁醇和水层萃取部位的出膏率分别为24.4％、8.20％、4.92％、11.47％和19.67％.新疆假龙胆乙醇提取物、水萃取部位、正丁醇萃取部位、乙酸乙酯萃取部位组小鼠经口给药的最大给药量分别为86.88、203.35、348.73、325.20 g生药/kg体质量.结论 新疆假龙胆乙醇提取物及其不同极性萃取部位均安全无毒.
