万古霉素对金黄色葡萄球菌生物膜黏附能力的影响

         

摘要

目的 探讨万古霉素对金黄色葡萄球菌形成生物膜早期黏附能力的影响.方法 孵育金葡菌在玻片上形成生物膜,对玻片上形成的细菌胞外多糖(extracellular polymeric substances,EPS)用荧光增白剂染色、黏附的细菌用SYTO9 /PI染色,再用激光共聚焦显微镜观察结合Image-Pro Plus Version 6.0(IPP)图像分析软件对各组EPS参数和细菌菌落进行定量分析.结果 加入万古霉素6小时后,1/2最低抑菌浓度(minimum inhibitory concentration,MIC)、1MIC、2MIC和4MIC组的EPS的面积和累积光密度(integrocted optical density,IOD)值分别为2183.33±923.90和3510.56±1482.10、3364.00±1885.30和5400.09±3026.40、2998.33±427.70和4815.24±689.10、3908.33±2030.70和6273.83±3259.90,均比空白对照组明显增加,P<0.05;1/2MIC、1MIC和2MIC组的菌落变密,P<0.05.结论 万古霉素加入到培养液后,使金葡菌黏附在玻片上的数量增多,形成的多糖基质面积增大,细菌分泌多糖基质时间提前,表明在金葡菌生物膜形成的早期黏附阶段,万古霉素能促进金葡菌胞外多糖基质的形成,促使金葡菌与材料表面的粘附能力增强.%Objective To investigate the effects of Vancomycin on the adhesion ability of the staphylococcus aureus. Methods Staphylococcus aureus were grown to biofillns on the glass. The extracellular polymeric substanees(EPS) of staphylococcus aureus was stained with the fluorescent whitening agent,the adherent bacteria was stained with SYTO9/PI. The laser scanning conffocal microscope in combination with image-Pro Plus Version 6.0software was used to perform the quantitative analysis of the EPS and the bacterial colony. Results Six hours after adding Vancolnycin,the area and IOD value of EPS of 1/2MIC ( lninimum inhibitory concentration) , 1MIC,2MIC and 4MIC were 2183.33 ± 923.90 and 3510.56 ± 1482.10,3364.00 ± 1885.30 and 5400.09 ± 3026.40,2998.33± 427.70 and 4815.24 ± 689.10,3908.33 ± 2030.70 and 6273.83 ± 3259.90 respectively. They were all significantly increased after intervention with vaneolnyein. The bacteria in the EPS increased and colony closed up after administration of 1/2MIC, 1MIC and 2MIC concentration(P < 0.05 ). Conclusion The EPS and the bacteria increased after intervention with Vaneolnyein. Vaneomyein can increase the initial adhesion ability of staphylococcus amreus.
