Some chitosan films were fabricated by casting method using glycerol as plasticity agent in dilute acetic acid solution and then characterized.Rat dermal fibrobalasts and human neofetus dermal fibroblasts were cultured in vitro to evaluate the biocompatibility of the chitosan films.The results showed that some cells cold be broken at early stage of the culture because there were too many basic amino groups in chitosan which would tightly bind the negative surface of cells.However,while the culture was continued,cells could normally grow and proliferated.It was found that new cells would be inhibited by contact to each other after they were spread all over the surface of the films.In addition,it was also observed that human neofetus dermal fibroblasts grew slower than rat dermal fibroblasts.%为考察壳聚糖对皮肤成纤维细胞的相容性,采用流延法制备了纯壳聚糖膜和含甘油的壳聚糖膜,并在所制备的壳聚糖膜上进行了大鼠皮肤成纤维细胞和人胎儿皮肤成纤维细胞的体外培养.结果表明,壳聚糖膜在培养开始阶段会导致部分贴附细胞的破坏,而进一步培养会促进这些细胞的生长和增殖.当细胞长满膜表面发生互相接触时,细胞产生接触抑制现象,不再继续增殖.研究还发现,不论在标准培养板上还是在不同的壳聚糖膜上,人胎儿皮肤成纤维细胞的生长速率均小于大鼠皮肤成纤维细胞的生长速率.
