CETP TaqIB基因多态性与血脂及冠心病相关性研究

         

摘要

目的:探索CETP TaqIB基因多态性与脂代谢及冠心病(CHD)发生的相关性。方法:选择天津市胸科医院住院病人430例,分为CHD组322例,对照组108例。入院后测定总胆固醇(CHO)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、载脂蛋白A1(Apo-A1)、载脂蛋白B(Apo-B)水平。采用PCR-RFLP方法测定研究对象CETP TaqIB基因型,分析CETP TaqIB基因多态性与血脂水平及冠心病发生的相关性。结果:(1)在CHD组,CETP TaqIB基因型B1B2、B2B2的HDL-C水平[(1.29±0.35) mmol/L、(1.16±0.36) mmol/L]较基因型B1B1[(1.09±0.22) mmol/L]相比有增高的趋势,但无显著的统计学差异(P>0.05),CHO、TG、LDL-C、Apo-A1、Apo-B水平均无显著差异(P>0.05);对照组基因型间血脂水平无显著差异。(2)B1B1、B1B2、B2B2基因型频率在CHD组为35.1%、45.0%、19.9%,在对照组为37.0%、43.5%、19.5%;B1、B2等位基因频率在CHD组及对照组分别为57.6%、58.6%和42.4%、41.2%。两组CETP TaqIB基因型及等位基因分布无显著统计学差异(P>0.05)。结论:天津地区人群CETP TaqIB等位基因B2频率分布与国内其他地区人群相一致;尚不足以证明CETP TaqIB基因多态性与冠心病存在相关性。%O bjective:To investigate the correlation between CETP TaqIB gene polymorphism and lipid metabolism and coronary heart disease (CHD). Methods:A total of 430 patients in Tianjin Chest Hospital were selected in the analysis. All the enrolled patients were divided into CHD group (n=322) and the control group (n=108). After admission total cholesterol (CHO), triglyceride (TG), high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), apolipoprotein A1 (ApoA1), apolipoprotein B (ApoB) were tested. PCR-RELP was executed to determine the genotypes of CETP TaqIB. Results: (1) In the CHD group, the CETP TaqIB genotype B1B2,B2B2 levels of HDL-C [(1.29±0.35) mmol/L, (1.16±0.36) mmol/L] were higher than genotype B1B1 level of HDL-C [(1.09±0.22) mmol/L], but no statistically significant difference (P>0.05) was found. The levels of CHO,TG,LDL-C,Apo-A1,Apo-B were statistically significant. No significant difference was found in the level of lipid in the control group genotype. (2) The frequencies of genotype B1B1, B1B2 and B2B2 were 35.1%, 45.0%, 19.9%in CHD group, and 37.0%, 43.5%, 19.5%in the control group. The frequencies of B1, B2 allele were 57.6%, 58.6%in CHD group, and 42.4%, 41.2%in the control group. No statistical significance in genotype and allele frequency was found between CHD group and the control group (P>0.05). Conclusion:CETP TaqIB gene B2 people in Tianjin area is consistent with other parts of the country. The existence of correlation between CETP TaqIB gene polymorphism and coronary heart disease needs further evidence.
