骨形态发生蛋白-1在盆底器官脱垂患者阴道组织中的表达及意义

摘要

Objective:To analyze the expression profile of bone morphogenetic protein-1 in vaginal tissue of patients with pelvic organ prolapse and normal controls. Methods: 15 patients with pelvic organ prolapse were recruited in this study,as well as 8 patients without SUI or POP as controls. Total mRNAs of BMP-1 were quantified by real-time RT-PCR and expressions of BMP-1 -1 (70 kDa)and BMP-1-3(130 kDa) isoforms were detected by Western blotting. Results:Bone morphogenetic protein-1 gene decreased in postmenopausal pelvic organ prolapse patients compared with asymptomatic women (P <0. 01). The expression of 130 kDa isoforms of BMP-1 were down-regulated in postmenopausal patients (P<0. 05), whereas no statistical difference of the 70 kDa isoform expression was found between the two groups. Conclusions: The bone morphogenetic protein-1 expression was down-regulated in the postmenopausal patients with pelvic organ prolapse. Dysregulation of BMP-1 may contribute to dysfunctional pelvic connective tissues.%目的:探讨骨形态发生蛋白-1(BMP-1)在盆底器官脱垂患者阴道组织的表达情况及其在盆底器官脱垂发生中的作用.方法:选择南京军区福州总医院收治的绝经前盆底器官脱垂患者15例为研究组,同期选取因妇科良性疾病行全子宫切除8例患者为对照组.于术中取患者阴道前壁组织,采用RT-PCR检测BMP-1 mRNA的表达,采用免疫印迹方法检测BMP-1两个主要表型(BMP-1-1、BMP-1-3)蛋白表达.结果:盆底器官脱垂患者阴道前壁组织中BMP-1 mRNA以及BMP-1-3蛋白的表达与对照组比较明显减低,差异有高度统计学意义及统计学意义(P＜0.01,P＜0.05)；而BMP-1-1蛋白的表达在两组之间比较,差异无统计学意义(P＞0.05).结论:盆底器官脱垂患者BMP-1表达降低,从而使成熟胶原释放减少,胶原蛋白含量降低,这可能是盆底器官脱垂的发生机制之一.
