Identification of a Core Sequence Within 5' Regulatory Region of Porcine Cytochrome P-450 1A1 Gene

Cytochrome P-450, subfamily I, polypeptide 1 (CYP1A1) encodes a member of cytochrome P450 superfamily of enzymes. This protein localizes to the endoplasmic reticulum and its expression is induced by some polycyclic aromatic hydrocarbons (PAHs), some of which are found in cigarette smoke. To understand the core sequence within the 5' regulatory region of porcine CYP1A1 gene, nine primer pairs were designed to amplify the different length of the CYP1A1 5' regulatory region, then the PCR products were constructed to the Firefly luciferase gene report vector PGL3-basic. Detection results in PK15 cell and fibroblasts all showed that a 39 bp fragment (-68 bp--29 bp upstream) was the core sequence of porcine CYP1A1 promoter. Further research found that HSF and SRY might be the most important transcriptional factor for porcine CYP1A1 promoter.
