亚硝酸诱变对火木层孔菌的影响

         

摘要

Objective The study of mutagenic treatment on Phellinus igniarius was carried in order to improve the content of secondary metabolites, such as polysaccharides and flavonoids, and enhance pharmacological activities. Methods The nitrous acid mutagenesis was chosen to mutagenic treatment of P. igniarius, and the relationship of nitrous acid concentration and mutagenesis time with death rate was investigated. The differences of the representative strains PNCM5-1 after mutagenesis by 0.005 mol·L-1 nitrite and P. igniarius on polysaccharide content and flavonoid content were compared. Results Taking the stability of fungi and the pigment secretion of secondary metabolites as detecting index, the best process time was 60 minutes on0.005 mol·L-1 of nitrous acid, and 8 minutes of 0.25 mol·L-1, respectively. The dominant strain PNCM5-1 reached the secondary metabolic process earlier and its production of flavonoids was higher than that of the original strain P. igniarius before 12 days' culture. Conclusion The strain of P. igniarius was regulated by nitrous acid mutagenesis and its culture reached the secondary metabolic process earlier than P. igniarius.Therefore, this could save the culture time for the production of flavonoids and provide a basis for the further development and utilization of the active ingredients of medicinal P. igniarius.%目的 对火木层孔菌进行诱导改造以提高该种菌的多糖和黄酮等次级代谢产物含量.方法 采用亚硝酸诱变方法对火木层孔菌菌株进行诱变处理,研究两种诱变浓度下亚硝酸的诱变时间与火木层孔菌致死率的关系,比较火木层孔菌与0.005mol·L-1亚硝酸诱变后代表菌株PNCM5-1的代谢产物在多糖含量和黄酮含量方面的差异.结果 以诱变菌株的稳定性和代谢产物颜色变化为考察指标,确定浓度0.005mol·L-1与0.250mol·L-1亚硝酸的最佳诱变时间分别为60和8min.优势菌株PNCM5-1比原始菌株先到达次级代谢过程,黄酮类化合物的产量在培养12d之前均高于原始菌株.结论 通过亚硝酸诱变,菌株的代谢受到调控作用,诱导菌株优先到达次级代谢过程,为桑黄生产黄酮类化合物节省培养时间,为药用桑黄活性成分的进一步开发与利用提供基础.
